| Objective:To explore the role of phosphorylation IKK and phosphorylation IκBαin the development of diabetic nephropathy(DN)in diabetes mellitus rats.And to offer a proof for blocking NF-κB path.Methods:64 Wistar rats were divided randomly into two groups.32 rats in normal control group and 32 rats in diabetes mellitus group.The subjects in diabetes mellitus group were injected intraperitoneally with dose STZ 50mg/kg while the ones in normal control group were injected intraperitoneally with the similar dose of citric acid buffer.At 1,4,8 and 12 weeks, collecting 24-hour urinary,paralyzing and obtaining blood,weight,fast Blood glucose,24-hour urinary microalbumin were observed,serum urea nitrogen,creatinine was measured,the kidney was fixation and embedment,the pathological changes of kidney were examined using light microscope and electron microscope.Renal expression of phosphorylation IKK and phosphorylation IκBαwere determined by immunohistochemistry.Results:Compared with the control group,diabetic rats expressed more 24-hour urinary microalbumin quantitation was significantly higher than the control group(P<0.01)at 4weeks,8weeks and 12weeks;serum urea nitrogen,creatinine were not significantly higher than the control group;pathohistological study revealed that the volume of glomerule of diabetes mellitus group was larger in the early stage,there was thicker GBM in diabetic rats(P<0.01),the renal tubular epithelial cell vacuolar degeneration,immunohistochemisty showed that the expression of phosphorylation IKK in renal tissue was much higher than normal control group(P<0.01), phosphorylation IκBαalso more expression than the control group(p<0.01),they increased continually during the whole experiment,There was a significant positive correlation between the expression of phosphorylation IKK and phosphorylation IκBαin renal tissue of DM group at 1,4,8 and 12weeks(r=0.979,P<0.01),between urinary microalbumin and phosphorylation IKK(r=0.8898,P<0.01),and between urinary microalbumin and phosphorylation IκBαin renal tissue of DM group at1,4,8 and12 weeks(r=0.878,P<0.01).Conclusion:The phosphorylation IKK in diabetic group expressed in renal tubular epithelial cell and glomerulus endotheliocyte endochylema,the expression was more stronger, complying with the progress of diabetes mellitus,with the activation of IKK,we can observed more phosphorylation IκBαexpression in the renal tissue endochylema。Associating with renal pathohistological and biochemical indicator,we can make a conclusion:in the progress of diabetic nephropathy,IKK and IκBαplay a positive role. |
