| Objective:To investigate relationship between the expression ofTGF-β1,Smad4 and VEGF and the recurrence or metastasis of colorectalcarcinoma,and to analyze combined detection of TGF-β1,Smad4 andVEGF on estimating the prognosis of colorectal carcinoma.Methods:Selecting 60 cases of colorectal carcinoma from The Third XiangyaHospital and Hunan Tumor Hospital between 2001and 2004 andarranging these samples into the recurrence and metastasis group and thedisease free survival group with clinical metastasis and metastasis or notwithin 2~5 year.All patients,with complete clinical data,were confirmedby pathology and surgery. Patients in both of groups were received noradiotherapy,chemotherapy or immunologic therapy before operation.The expression of TGF-β1,Smad4 and VEGF were detected with SPimmunohistochemistry. Combining with the clinical survival data, all thedata were analyzed by the software of SPSS.13.0 for windows toinvestigate the effect of TGF-β1,Smad4 and VEGF on predicting therecurrence and metastasis of colorectal carcinoma.Results:1. Compared with less than or equal to 3cm group, The expression ofTGF-β1,Smad4 and VEGF in greater than 3cm group was significant difference respectively(P<0.05). Compared with poorly differentiatedgroup, The expression of TGF-β1,Smad4 and VEGF in well-moderatelydifferentiated group was significant difference respectively (P<0.05).Compared with Dukes A or Dukes B stage group, The expression ofTGF-β1,Smad4 and VEGF in Dukes C or Duke D stage group wassignificant difference respectively (P<0.05).2.The expression of TGF-β1 in recurrence and metastasis colorectalcarcinoma tissues were significantly higher than that in no-recurrence andno-metastasis colorectal carcinoma tissues (p<0.05).The expression ofSmad4 in recurrence and metastasis colorectal carcinoma tissues weresignificantly lower than that in no-recurrence and no-metastasis colorectalcarcinoma tissues (p<0.05).The expression of VEGF in recurrence andmetastasis colorectal carcinoma tissues were significantly higher than thatin no-recurrence and no-metastasis colorectal carcinoma tissues (p<0.05). The expression of Smad4 in TGF-β1 positive expression colorectalcarcinoma tissues was significantly lower than that in TGF-β1 negativeexpression colorectal carcinoma tissues (p<0.05).Conclusion:1..The up-regulated expression of TGF-β1 and the down-regulatedexpression of Smad4 in the recurrence and metastasis group indicateschange of the TGF-β1 signal transduction pathway in colorectalcarcinoma, might be concerned with the metastasis and recurrence of colorectal carcinoma.2. The higher expression of TGF-β1, VEGF proteins and a lowerexpression of Smad4 protein in the recurrence and metastasis group,which indicates alterations of TGF-β1, Smad4 and VEGF gene may playan important role in the metastasis and recurrence of colorectalcarcinoma.Combining detecting the expression of TGF-β1,Smad4 andVEGF proteins in colorectal carcinoma is helpful to our understanding ofclinicopathological features and behavior, and could be importantpathological biology index to predict the recurrence,metastasis andprognosis of colorectal carcinoma. |
