Observation Of The Curative Effect After Transfering Endostatin Gene To Endometriosis Rat Model

Objective: To establish the endometriosis rat models, injectingendostatin gene to the ectopia focus, analysis of the changes of endostatin,vascular endothelial growth factor,matrix metalloproteinases-2,volum andmicrovessel density, oberserve the growth of ectopia focus after thistherapy.Methods: The study used 85 female Lewis rats of 14 weeks andunpregnancy to establish the endometriosis animal models bytransplanting an autologous fragment of endometrial tissue onto the innersurface of the abdominal wall. To select the 64 succeeded rats after 4weeks, dividing them to three groups randomly, one group for transferingendostatin gene (24 rats), one group for blank plasmid(20 rats), and onegroup for PBS solution(20 rats). The level of the endostatin and vascularendothelial growth factor in serum were detected by ELISA, theendostatin, MMP-2 and microvessel density(MVD) in ectopia endometriawere detected by immunohistochemical assay, and the volume of theectopia focus were detected by vernier cursor.Results: After transfering endostatin gene, the level of the ES andVEGF in serum were detected by enzyme-linked immunosorbentassay,the difference between the group for transfering endostatin gene and thetwo control groups have statistical significance, the level of ES isincreased(P＜0.01), and the level of VEGF is decreased(P＜0.01), therewas an negative correlation between them in serum(r=-0.805). Theexpression of the ES,MMP-2 and MVD in ectopia endometria weredetected by immunohistochemical assay, the difference between thegroup for transfering endostatin gene and the two control groups havestatistical significance, too, the expression of ES is increased(P＜0.01), the expression of MMP-2 is decreased(P＜0.01), and there was an negativecorrelation between them in ectopia endometria (r=-0.700), the MVD isdecreased(P＜0.01), The volume of the ectopia focus were detected byvernier cursor, the difference between the group for transfering endostatingene and the two control groups have statistical significance, the volumeof the group for transfering endostatin gene is smaller thanbefore(P＜0.01), and the volume of the two control groups are bigger thanbefore(P＜0.01).Conclusions: Injecting the lipofectamine-endo-pBud into ectopicfocus has definite curative effect in endometriosis rat model.
