| Flavonoids,which the basic structure being C6-C3-C6, is the secondary metabolic substance,and exist in the natural plant in the form of dissociation state or glucoside. Not only are flavonoids quantity and sort various, but also their structure type are complicated. These characteristics determaine the activities of flavonoids on respiratory system disease, heart and brain blood-vascular disease, such as anti-inflammation, restraining fungus, anti-oxidant, anti-radiation, anti-tumor and strengthening immune ability, etc. In order to explore the abundant resources of pteridophyte and look for the effective chemical component on the protective effects of skin cells.So it is necessary to extensive study about extraction, isolation , identification and biological and medicine activities of flavones form pteridophyte roots.This study is composed of two parts. Part I, The purpose of our study was the optimum extracting methods,isolation and identification of flavonoids form pteridophyte roots.Part II mainly introduces biological activities of flavonoids form pteridophyte roots by investigate the effects of flavonoids to mouse skin cells.First,The purpose of our study was to get a kind of natural component total flavonoids from pteridophyte roots, we collected abundant pteridophyte roots, we investigated the optimum extracting condition of flavonoids from pteridophyte roots by using the gorthogonal design method. And the results show temperature 80℃, 70% ethanol , extracting 6h were optimal. In this condition, we extracted the flavonoids and measured its content. The result indicated the average content is 1.26%.Second,we used three ways to identificated the flavonoids from pteridophyte roots. The three ways separately were ultraviolet spectrum and TCL methed.Tird, mouse skin cells culture in vitro. Third passage of mouse skin cells were investigated for the effect of flavonoids on the proliferation capacity of mouse skin cells. Cultured cells were placed in 96-well culture plates and incubated for 24 hours,then treated with various concentrations of flavonoids (100 -800μg/mL).Cultured cells were investigated by MTT assay after exposure to the drugs 48 hours .Flavonoids from pteridophyte roots has a significant effect on the proliferation of mouse skin cells. The effect is in a dose-dependent tendency,600μg/mL flavonoids is a most effect on proliferation. To sum up, the new discovery of this research are as follows:This study provides a scientific basis for extracting,isolating,purificating, identifying on flavonoinds of pteridophyte roots. Biological and medicine activities of flavonoids form pteridophyte roots has been synthesized by cytobiology technology. The experiment provides a scientific basics for selection of drugs to protective foundation of skin cells. |
