Effect Of Ginseng-Schisandrae Decoction On IL-4, IFN-γ In BALF And MMP-9, TIMP-1 In Lung Tissue From Asthmatic Mice

Objective: To study the therapeutical effect and mechanism of Ginseng-Schisandrae Decoction on asthmatic mouse Methods: 40 mice were randomly divided into five groups averagely: negative control (A), positive control (B), low dose Ginseng-Schisandrae Decoction group (C), large dose Ginseng-Schisandrae Decoction group (D), and Budesonide group (E). The mouse models of asthma were made by the egg albumen sensitized method and were treated with low dose, large dose Ginseng-Schisandrae Decoction and Budesonide for four weeks, Differential cell counting were counted from bronchoalveolar lavage fluid(BALF) and histological changes of the lungs were evaluated after HE stain. Interleukin-4 and Interferon-γin BALF were assessed with ELISA, the expression of Matrix Metalloproteinases -9 and Tissue Inhibitor Metalloproteinase -1 in lung tissue were examined with immunohistochemical method. Result①Gasp and the lung inflammation of the mice from C, D, E groups were significantly lessen than those from B group, and A group is normal.②Total cells, eosinophils, Neutrophil, and Lymphocyte in BALF from A, C, D, E groups were significantly decreased compared with those from B group (P<0.01) as well as the same changes with A, E groups to C, D groups (P<0.01).③In A, C, D, E groups, Interleukin-4 and Interleukin-4/Interferon-γwere decreased and Interferon-γwas elevated when compared with B group (P<0.01), furthermore, there was also significant different between A, E groups and C, D groups (P<0.01).④Matrix Metalloproteinases -9, Tissue Inhibitor Metalloproteinase -1 and their ratio in A, C, D, E groups were evidently lessen than those from B group (P<0.01). there was also significant different between A, E groups and C, D groups about Matrix Metalloproteinases -9 and Tissue Inhibitor Metalloproteinase -1 (P<0.01 or P<0.05). all of the parameters had no difference between A, E and C, D groups. Conclusion: Ginseng-Schisandrae Decoction could treat bronchial asthma, the possible mechanism is that Ginseng-Schisandrae Decoction could down-regulate Interleukin-4, Matrix Metalloproteinases -9, Tissue Inhibitor Metalloproteinase -1, heighten Interferon-γand regulated the balance between Interleukin-4, Interferon-γ, and Matrix Metalloproteinases -9 Tissue, Inhibitor Metalloproteinase -1.
