Objective To investigate the expression of HIF-1a,HPSE and their correlation withvascular endothelial growth factor and microvessel density in retinoblastoma.Methods HIF-1a,VEGF was detected by SP immunohistochemical method in 34cases of retinoblastoma and 10 cases of normal retina tissues, and microvessel density(MVD) was measured by anti-CD34. Eighteen samples of retinoblastoma tissues andten samples of normal retina were examined for HIF-1a mRNA,HPSE mRNA,VEGFmRNA expression by RT-PCR. HIF-1a,VEGF,HPSE expression were analysed withtheir relation to microvessel and clinical,pathological character.Results The positive expression rate of HIF-1a,HPSE and VEGF were higher inretinoblastoma than in normal retina's. The expression of HIF-1a and VEGF inretinoblastoma were correlated with MVD. VEGF was correlated with HIF-1a and HPSEexpression.Conclusion HIF-1a and HPSE have effect on the expression of VEGF. HIF-1a,HPSEand VEGF play a role in tumor angiogenesis and promote malignant progress ofretinoblastoma.
