Objective: to provide theoretical evidence that transplanting neural stem cells can treat epilepsy, we explore the survival and differentiation and migrating of neural stem cells of GFP(Green fluorescent protein)transgenic mice in brain of epileptic rat, and test the effect of the cells to the apoptosis in the epileptic rats. At present, there is no report about implanting stem cells to treat epilepsy.Methods: penicillin was used to induce the rat models of epilepsy by intraperitoneal injection. The rat model of the control group and the study group were established.The control group was divided into two groups, the normal control group(N=8)and the epileptic control group(N=8).The study group was divided into two groups,the normal saline group (N=24) and neural stem cells (N=24) managed by transplanting.Neural stem cells were isolated and cultured in vitro.Then normal saline,the neural stem cells suspension were grafted into the right hippocampus of the rat models of EP through orientation. After transplantation 7 days, 14 days, 28 days, fluorescence microscope was used to observe the survival of the transplanted neural stem cells, immunohistochemical method was used to test the apoptosis gene Bax, Bcl-2 protein, electroencephalogram was used to monitor electrical activity of brain of the rat.Results: we found that the transplanted neural stem cells can survive and migrate in the brain of epileptic rat, These cells also can integrated with the peripheral tissue, cell hyperplasia couldn't been found. The number of the survival cells at 14 days after grafting was less than its at 7 days(P<0.01),the number at 28 days was less than its at 14 days(P<0.01).The number of the survival cells decreased gradually along with the time gone.The number of the apoptosis cells in the epileptic hippocampus of the neural stem cells groups were less than the normal saline groups after transplantation 7days, 14days and 28 days respectively (P<0.01). Meanwhile with the time gone, transplantation of stem cells can dramatically alleviated seizure activity in EEG recordings and obviously reduced the amplitude of discharge.Conclusions: our results demonstrate that neural stem cells transplanted into the hippocampus of the rat models of EP through orientation can survive and migrate, dramatically alleviate seizure activity in EEG recordings. In some certain these cells can depressed the cells apoptosis of the hippocampus regions resulted from epilepsy. It may be concerned with the repair the brain of epileptic rat.It suggests that cell grafting may be a valid means for curing epilepsy.
