ObjectiveAfter cataract is after extracapasular cataract or damaged the lens leading the posterior capsule opacification. Recently ,as we known,the LECS alived after cataracr operation prouduceing,moving,fibrosis cause the PCO. With the operation technology enhanced and the operation equipment improved,the following symptom after cataract operation is less and less.But PCO is alson the common symptom,especially in children.The rate of PCO is about 100%,which is badly influencing the forward sight.Recently although many foreigners using many ways to prevent PCO,it also not soluted.Using animal experiment to investigate the inhibitive effect of elemene pouring into anterior chamber after extracapasular cataract extraction on posterior capsule opacification in rabbit.MethodsChoosing 40 healthy rabbits, the bady weight is 1.5-2.0kg, 2-4months old.These rabbits are stochasis into 5 groups, which are comparisonal group and experimental groups. The experimental groups are formed the different concentration elmene ,which are 100ug/L,200ug/L,300ug/L,500ug/L,8rabbits in each group.After the extracapasular cataract extraction, elemene with different concentration (100ug·L -1 200 ug·L-1 300 ug·L-1 500 ug·L-1) was poured into anterior chamber after extracapasular cataract extraction for 5min , then using BBS instead of it for 10min.The followed time after operation was 3 months the PCO and the change of intraocular pressure were observed and compared between control group and experimental group.Count cornea endothelium density before and after operation, and check the cornea endothelium shape.Results1 the degree posterior capsule opacification after operationAt the 2 weeks after operation the comparisonal group appear PCO, at the 4 weeks after operation ,the 100ug/L group appear PCO , at the 7 weeks after operation ,the 200ug/L group appear PCO , at the 9 weeks after operation ,the 300ug/L group appear PCO , at the 10 weeks after operation ,the 500ug/L group appear PCO .At the 2 weeks after operation , the PCO only appeared in the control group. 12 weeks after operation , the PCO in the eyes of experimental group was not so obvious as that of control group , and higher the concentration of the drug was, the less PCO showed. The cornea endothelium density there was obvious different from control group in 500ug·L-1.2 counting the corneal endothelium at 3d 7d 15d 1month after operationAt 3d 7d 15d 1month after operation ,the experimental groups corneal endothelium cells are different from comparisonal group(.P<0.05).Using multiple comparisons 100ug/L,200ug/L,300ug/L is same as the comparisonal group(P>0.05).The 500ug/L is different from comparisonal group(P<0.05).3 The change of intraocular preesure before and after operationUsing multiple comparisons ,before operation and 3 days after operation are different from 1 and 2 days after operation,(P<0.05) before operation is same as 3 days after operation(.P>0.05).4 The change of ERGb wave before and 15,30 days after operationThe 15,30 days is same ascomparisonal group (P>0.05). ConclusionElemene can inhibit the proliferation of lens epithelial cells effectively , which is related to the concentration of drug.Elemene applied in the anterior chamber with the concentration of 100-300 ug·L-1 is safe and effective in inhibiting the PCO.
