Supercritical fuid(SCF) is becoming research interests gradually since it had been discovered, and its relative studies is increasing. Supercritical CO2, with the advantages of innocuity, no pollution, low viscosity, high diffusion coefficient, and easy obtainability, had been applied widely, such as drug extraction, purification, particulatization and pharmaceutical impregnation, pharmaceutical impregnation, which could obtain slow-released drug to prolong drug effect time and reduce dose time,was the key study focus. However, a main issue in the study was qualitative and quantitative detection, which was to get relevant data, and provide good sustainment for production and application.There are two main detection ways at present, on-line detection and off-line detection. On-line detection could respond truly to reaction, and was an important developing direction. The on-line detection had many methods, such as weigh, volume and spectrum detection. All the methods needed proper reaction device, and researching and developing a dependable on-line detection device was a very important field nowadays.The current status of pharmaceutical impregnation and its detection was analyzed and reviewed in this dissertation, with deficiency and developing direction pointed out. The main work was developing a high-temperature high-pressure reaction device independently based on nowaday research. The device could adjust the reaction accurately and detect the reaction on line, which established a base for future relative study. At the same time, the concept of non detection of multi- nonlinear non-balance complex system was proposed, and the advantage and importance of its implication in the study was illustrated. The dissertation contained the following parts. The status of current pharmaceutical impregnation and its on-line detection was reviewed in the first part, and the relative issues was analysed. The second part introduced the reaction principle and device design principle. The third part mainly introduced the reaction device developed independently. The fourth part introduced the common detection methods, the concept and research meaning of complex system; the fifth part was the summarization and the research prospect.
