Due to deterioration in ovarial function and decreased hormone secretion-especially estrogenic hormone, which leads to great imbalance of gonadal hormone and metabolism of glucolipid in body, form adipose tissue. women in perimenopausal period are at higher risk for developing chronic diseases, such as heart disease, diabetes and hypertension. In Traditional Chinese Medince, this manifestation falls into the category of 'pre-/post- menopause syndrome'. Its pathogenesis is that of obesity. Huangdi's Internal Classics notes that it is associated with the levels of QI and blood as well as phlegm turbidity and blood stagnation in the body. So Salvia miltiorrhiza has the function of activating blood circulation to dissipate blood stasis, However, effects of Salvia miltiorrhiza on ovariectomized obese rats remain unclear.To simulate metabolism of female in perimenopause and postmenopause ovariectomized(OVX) rat model was made in the experiment. Body mass, food intake, water intake, Lee's Index were measured, and serum levels of blood lipids, estradiol(E2), blood glucose, insulin(Ins), glutamate pyruvate transaminase(GPT), glutamate oxalacetate transaminase(GOT), malondialdehyde(MDA), total antioxidant capacity(T-AOC), Leptin, interleukin-6(IL-6), tumour necrosis factor-a(TNF-a) were assayed in the experiment. The effects of Salvia miltiorrhiza in ovariectomized obese rats and its mechanisms involved were investigated.Twenty-four healthy female SD rats were randomly divided into three groups with 8 rats in each group, namely shame-operated group(SHAM), ovariectomized(OVX) group and Salvia miltiorrhiza(SM) group. After having been narcotized by ketamine(100mg/kg), rats were underwent a bilateral ovariectomy except those in the sham-operated group which were also operated a bilateral fat as much as ovariectomized group, but their ovaries were not cut off. Rats in shame-operated group and ovariectomized group freely drank water. Rats in Salvia miltiorrhiza group freely drank 1% water extracts from Salvia miltiorrhiza postoperatively, later the concentration of Salvia miltiorrhiza gradually increased to 12%, which lasted until the end of the experiment. The food intake was monitored daily, and body mass was measured every five days. At the end of the experiment, all the rats were fasted for 12 hours, measured the stem length, tail length and blood glucose. collected blood samples for the measurement of blood lipids, estradiol(E2), blood glucose, insulin(Ins), glutamate pyruvate transaminase (GPT), glutamate oxalacetate transaminase(GOT), malondialdehyde(MDA), total antioxidant capacity(T-AOC), Leptin, interleukin-6(IL-6) and tumour necrosis factor-a(TNF-a). SPSS13.0 software was used and the data was statistically analyzed and the results were expressed as Mean±SD. The One-Way analysis of variance (One-Way ANOYA) was used for comparison among the three groups. PO.05 represented significant differences. The main results were as follows:1. Compared with SHAM group, Rats in OVX group displayed increased body mass(P<0.001), food intake(P<.05-0.001), Lee's Index(P<0.001) and serum level of blood lipids , glutamic pyruvic transaminase(GPT), glutamic oxalacetic transaminase(GOT), malondialdehyde(MDA), Leptin, tumour necrosis factor-α(TNF-α) (P<0.05-0.001), and reduced sernm level of estrogen(P<0.05-0.001) .however, water intake, blood glucose, insulin(Ins), total antioxidant capacity(T-AOC), nterleukin-6(IL-6) were no significant difference .2. Compared with OVX group, in SM group body mass(P<0.05-0.01), food intake(PO.05-0.001) and serum level of malondialdehyde(MDA) (PO.05), Leptin(P<0.05), tumour necrosis factor-a(TNF-a) (P<0.05) were lower, however, water intake, Lee's Index, blood glucose, insulin(Ins), total antioxidant capacity(T-AOC), glutamic pyruvic transaminase(GPT), glutamic oxalacetic transaminase(GOT), interleukin-6(IL-6) were no significant difference .The results indicate that Salvia miltiorrhiza can promote loss of body mass, reduce levels of triglyceride, malondialdehyde and tumour necrosis factor-a(TNF-a). increase sensitivity of leptin, and restrain the level of food intake in ovariectomized obese rats.
