Purpose To study the expression of survivin,Ezrin,interleukin-6 (IL-6) and matrix metalloproteinase-9 (MMP-9) in cervical carcinoma and their correlation with invasion and metastasis, seek possible tumor mark for cervical carcinoma; To search after what the survivin gene did during the cervical carcinoma, survivin mRNA were deteceted in vary pathology changes in cervix by in situ hybridization(ISH) at the same time.Methods The expression of survivin, Ezrin, IL-6 and MMP-9 in 40 cases of invasive cervical carcinoma, 38 cases of carcinoma in situ, 28 cases of cervical intraepithelial neoplasia, 15 cases of normal cervical epithelium was detected by immunohistochemistry SP method. The expression of survivin mRNA in 40 cases of invasive cervical carcinoma, 38 cases of carcinoma in situ, 28 cases of cervical intraepithelial neoplasia, 15 cases of normal cervical epithelium was detected by in situ hybridization.Results:1. survivin protein and survivin mRNA were significantly higher in invasive carcinoma compared to normal controls (P<0.05) . The positive rate of survivin protein in normal controls, cervical intraepithelial neoplasia, carcinoma in situ and invasive carcinoma were 0,53.6%,63.2%,80% resepectively. The positive rate of survivin protein in invasive carcinoma was higher than rest groups(P<0.05), the expression of survivin protein was not associated with age, clinic stage and histological grade, but was related to metastasis of lymph node. The positive rate of survivin mRNA in normal controls, cervical intraepithelial neoplasia, carcinoma in situ and invasive carcinoma were 0,42.8%,60.5%,72.5% resepectively. The positive rate of survivin mRNA in invasive carcinoma was higher than rest groups(P<0.05), the expression of survivin mRNA was not associated with age, but were related to clinic stage , histological grade, and metastasis of lymph node(P<0.05). The expression of survivin mRNA was positively related to that of protein (r_s=0.801, P<0.05).2. The expression of IL-6 was significantly higher in invasive carcinoma compared to normal controls and cervical intraepithelial neoplasia (P<0.05) . The strong positive rate of IL-6 in normal controls, cervical intraepithelial neoplasia, carcinoma in situ and invasive carcinoma were 46.7%,71.4%,81.6%,87.5% resepectively. The positive intension was significantly higher in invasive carcinoma than else groups(P<0.05),the expression of IL-6 was not associated with clinic stage and histological grade, but was related to metastasis of lymph node.3. The expression of MMP-9 was significantly higher in invasive carcinoma compared to normal controls and cervical intraepithelial neoplasia (P<0.05) , expression of MMP-9 was increased in carcinoma in situ compared to normal controls and cervical intraepithelial neoplasia (P<0.05) . The positive rate of MMP-9 in normal controls, cervical intraepithelial neoplasia, carcinoma in situ and invasive carcinoma were 33.3%,42.9%,60.5%,87.5% resepectively. the expression of MMP-9 was not associated with clinic stage and histological grade, but was related to metastasis of lymph node.There was a low correlation between IL-6 and MMP-9(r_s=0.462, P<0.01).4.The expression of Ezrin was significantly higher in invasive carcinoma compared to normal controls and cervical intraepithelial neoplasia (P<0.05 ) . The positive rate of Ezrin in normal controls, cervical intraepithelial neoplasia, carcinoma in situ and invasive carcinoma were 26.7%,64.3%,63.15%,80% resepectively. The expression of Ezrin was not associated with clinic stage and histological grade, but was related to metastasis of lymph node.Conclusions1. The high expression of survivin,IL-6,MMP-9 and Ezrin in cervical carcinoma might be correlated with invasive behavior of cervical carcinoma. They might become useful prognostic indicators for early cervical carcinoma.2. The survivin gene play an important role in the occurance and development of cervical carcinoma.There was no significance of the positive rate between survivin mRNA and survivin protein as a whole, It indicate that the regulation of the survivin protein maybe occur in the transcriptase level.3. There was a low correlation between IL-6 and MMP-9(r_s=0.462, P<0.01). in invasive cervicai carcinoma; The high expression of MMP-9 can be stimulated by IL-6, together construct a mico-envrioment of invasion and metastasis .Both correlated with invasive behavior of cervical carcinoma.
