| ã€Topic】An Experimental Study on Effects of BiXieHuaZhuoShuan(BXHZS) on TNF-αIL-1βand iNOS in Chronic Abacterial Prostatitis Rat Modelã€Objective】Chronic Prostatitis, a commonly encountered disease of male adult, is a clinical syndromes with the main effect of dysuric symptom and bladder genital area pain. As the lack of deeply researching to the pathogeny and the mechanism of the disease, effectiveness the etiology and pathophysiology of chronic prostatitis haven't been fully clarified in most cases, while recent studies indicated immunologic processes as a possibly major component to the pathogenesis of the disease, which has arisen enormous enthusiasm of an increasing number of researchers. Although a wide range of therapeutic approaches could be employed to chronic prostatitis, the actual effects remain unclear and dismal, resulting in the chronic, relapsing, and episodic nature of patients'symptoms. However, traditional Chinese medicine has shown considerable superiorities in managing such patients and has gained outstanding achievements. In this research, we prospectively observed a possibly experimental benefit from BXHZS in chronic abacterial prostatitis in rats as a new treatment option. Then we set the expression of cytokines TNF-α,IL-1βand iNOSas a clue to further discuss their roles in the potential pathogenesis of the disease from immunopathological perspectives by means of immunohistochemical assay. At last, we devised BXHZS or as intervention conditions to study its related effects on this rat model and analyze the possible mechanisms.ã€Methods】To observe the morphological and molecular biological peculiarities of the experimental autoimmune prostatitis rat. model made by SC purified prostate in twice with immune adjuvant. 90Wistar rats (male) were randomly divided into control group(n=15) and animal model group . Except control group, the animal model group was chronic prostatitis made by SC purified prostate in twice with immune adjuvant. 23 days later, It was randomly divided into chronic prostatitis group(n=15),high doses of BXHZS(n=15),low doses of BXHZS(n=15),moderate doses of BXHZS(n=15)and QianLieAnShuan(QLAS)(n=15).After treatment the 90 Wistar rats were detected TNF-αIL-1βcontent by ELISA. 90 cases of paraffin-embedded tissues from all the prostates of the rats included were detected for the expression of iNOS,means of immunohistochemical assay, which cells were considered positive whose membranes and/or plasm were stained brown yellow and all samples were expression intensity by image analysis system.ã€Results】 1.The expression of IL-1βin chronic prostatitis group was significantly higher(P<0.01)than that of control group. Significant differences of IL-1βcontent were noted between BXHZS groups,QLAS group and chronic prostatitis group(P<0.01);2.The expression of TNF-αin chronic prostatitis group were significantly higher(P<0.01) than that of control group. It is Significantly different between BXHZS groups QLAS group and chronic prostatitis group in TNF-αcontent(P<0.01);3.Compared with those of the control group, the iNOS expression decreased significantly in the BXHZS group,QLAS group(P<0.01)。ã€Conclusions】1.BXHZS had obvious effects on experimental chronic abacterial prostatitis.The principal mechanism of BXHZS on treating chronic prostatitis is related with decreasing the levels of proinflammatory cyokines in prostates(TNF-αIL-1β) and express of iNOS relieving leucocytes infiltration; 2.Treatment with BXHZS can inhibit the over-expression in experimental chronic abacterial prostatitis through inflammation inhibition and immune regulation; 3.Immune factors may play an important role in chronic abacterial prostatitis. |
