Approach chromosomal aberration in thyroid neoplasm initially. Method: 104 cases of paraffin embedment tissue benign and malignant thyroid lesion as control(including 45 case of papillary thyroid carcinoma, 31 cases of follicular thyroid carcinoma, 3 cases of medullary thyroid carcinoma, 10 cases of thyroid adenoma and 15 cases of nodular goiter) was collected. All of the samples were taken from the department of pathology of the first affiliated hospital, medical college of Shihezi University from 1998 to 2006 and all were diagnosed and classified by more than two pathological professors referring to WHO standard. Collect the thyroid affection fresh sample 17 case and 6 case peripheral blood. Immunohistochemistry was used to detect the expression of COX-2 and S100A4 in thyroid cancer and thyroid adenomas. Using the method of cell culture and G banding, observe chromosomal aberration in peripheral blood and fresh tissue.Results: By the Immunohistochemistry research, the expression rate of COX-2 and S100A4 in thyroid cancers were higher than those in thyroid adenomas (P<0.01), has dependability, especially with lymphatic metastasis. The expression and distribution of Ki-67 has difference between follicular thyroid carcinoma and thyroid adenoma. The rate of COX-2 and S100A4 expression has no dependability. The chromosome in the peripheral-blood lymphocyte of papillary thyroid carcinoma has no abnormality.Conclusions: The expression rate of COX-2 and S100A4 should help discriminating the thyroid neoplasm. COX-2,S100A4 and Ki-67 should help discriminating thyroid adenomas and thyroid cancer. Linking morphology, blood vessels distribute and immunohistochemistry stain, diagnosis of thyroid adenomas and thyroid cancer can be helped.
