Objective:To study the molecular pathogenic mechanism of predispose to hypertension ,hyperlipoidemia and obesity in Kazakan, we investigated the association between the C491T polymorphism ofβ2-AR gene and blood pressure ,lipid levels ( include triglyceride,total cholesterol, high density lipoprotein- cholesterol and low density lipoprotein- cholesterol levels) and body mass index (BMI) in Kazakan of Xinjiang.Methods: In this base-controled study, we recruited Kazakan subjects from the pastoral areas in Xinjiang through epidemiological investigation. Peripheral blood were collected from all recruited subjects. The DNA was extracted by means of a standard phenol-chloroform method. Then the genotypes ofβ2-AR was identified by Polymerase Chain Reaction -Restriction Fragment Length Polymorphism(PCR-RFLP) in all the subjects. All subjects were divided into two groups(hypertensives group and normotensives group) according to the Guideline for Prevention and Treatment of Hypertension in China(2005).The distribution of genotypic and allelic frequencies between hypertensives and normotensives were studied. All subjects were divided according to the Recommended Guideline for Prevention and Treatment for Dyslipidemia of WHO(1997).The relationship between the genotypes ofβ2-AR and TG,TC,HDL-C ,LDL-C levels were studied. All subjects were divided into three groups according to the standard of WHO about the measurement of normal, overweight and obesity(18.5kg/m2≤BMI<25kg/m2 and 25kg/m2≤BMI<30kg/m2 and BMI≥30kg/m2 respectively). Then the BMI levels between two genotypes, the lipid levels in different BMI groups, the distribution of genotypic and allelic frequencies among three groups were studied.Results:①In the population, the frequencies of the genotype CC,CT were 98.82% and 1.18%respectively,no TT was found .The genotypic and allelic frequencies were in Hardy-Weinberg equilibrium(χ2=0.018, P=0.893). These two genotypic frequencies were 97.92% and 2.08% respectively in essential hypertensives, while no CT in normotensives. The allelic frequencies of C,T were 98.96%,1.04% and 100%,0% in two groups and there is a significant differences between these two groups(P=0.040)。In addition, systolic blood pressure and diastolic blood pressure had no significant difference in the groups classified according to genotypes(P﹥0.05). Through analyze the data stratified with the gender, there is a significant differences between these two groups。In addition, systolic blood pressure has significant difference in the groups classified according to genotypes.②TG, TC, HDL,LDL levels were increased in CT but had no significant difference between these two genotypes in this population. The TC genotype had higher LDL-C level than CC genotype in female, and had significant difference between these two genotypes(P=0.038).But divided according to the standard announced by WHO, genotype CT and the frequency of T allele were associated with high LDL-C(P=0.031).③The genotypic frequencies and allelic frequencies were not significantly different among three groups classified according to BMI levels (P﹥0.05) and BMI levels had no significant difference in the groups classified according to genotypes(P﹥0.05),but the TG, TC levels had significant difference among three groups classified according to BMI, and with the increasing of BMI ,the TG and TC levels were raised(P=0.004,0.000 respectively ).Conclusion:①The C491T polymorphism ofβ2-AR gene may be associated with essential hypertension, especially in male。Which implicates that it is possible one of the susceptible factors of essential hypertension in Kazakan of Xinjiang.②the C491T genotype ofβ2-AR gene is associated with higher LDL-C level in this population,especially in female ,which suggest it may play an important role in abnormal LDL-C in female Kazakan of Xinjiang.③the C491T polymorphism ofβ2-AR gene may not be associated with overweight , obesity in Kazakan of Xinjiang, which implicates that it may not play an major role in the mechanism of overweight , obesity in Kazakan.
