Two-photon excitation fluorescence technology was used to study the kinetics of 5-ALA-induced Ppâ…¨in DHL (follicular lymphoma cells) cells. 5-ALA-induced Ppâ…¨localization in DHL cells was also studied via double-label fluorescence microscopy. By the use of semiconductor laser with output wavelength of 630nm, optimal experiment parameters such as laser power density and laser dosage were investigated forALA-PDT.At first, some basic knowledge about leukemia and recent method used in the therapy of leukemia were introduced, and methods of photodynamic purging of leukemia cell in vitro and the research development of vitro purging in leukemia cells were presented. Meanwhile, principle of confocal laser scanning microscope with the application in bio-medical field and the advantage of two-photon technology were also presented.Then, we tried to use the two-photon excitation fluorescence spectroscopy, in the lambda mode, specific fluorescence intensity of ALA-induced Ppâ…¨under various concentrations and different incubation time was measured, thus we obtained the kinetics of ALA-induced Ppâ…¨in DHL cells. In combination with fluorescence probes of lysosome, mitochondrial and endoplasmic reticulum, distribution of Ppâ…¨in DHL cells were also analyzed with the method of calculating the correlation coefficients.Furthermore, semiconductor laser with the output wavelength of 630nm was used to irradiate DHL cells which incubated in vitro and the optimal parameters such as: light power density and light dosage for ALA-PDT on DHL cells were investigated.Finally, the main content and results were summarized and the main innovations in the paper were shown, and a plan for the further study and research in the near future was also made.
