Experimental Study Of The Effect Of Combination Of Danshen Root And Milkvetch Root On Rats With FGR Induced By Smoking And Drinking

Objective To study the effect of combination of Danshen root and Milkvetch root on rats with FGR-induced by smoking and drinking from growth parameter, growth-faceors,and morphology, to find out the effect and mechanism of combination of Danshen root and Milkvetch root and to provide the important and scientific basis for the new treatment of FGR.Methods 1. The model of FGR rats was made by means of passive smoking and drinking . Some pregent rats were treated with combination of Danshen root and Milkvetch root injection, some were treated with L-arginine. The birth weight, the length and Poundera Index of all fetal rats and the rate of FGR in the five groups were measured and compared. To explore the effect of combination of Danshen root and Milkvetch root on FGR. 2. Immunohistochemical assay was performed to detect the IGF-I and IGF-IR expression on placenta . To explore the effect of combination of Danshen root and Milkvetch root injectionon on the synthesis and excretion of growth-factors of FGR rats. 3. The placenta of rats were examined by HE-stainsection. The tissue were assessed for morphological change with light microscope and electron microscope.Results 1. In the experimental group, the FGR rats'body weight, height were significantly lower than those in the control group(P<0.05). 2. In the FGR rats'placenta, the expressions of IGF-I and IGF-IR were decreased(P<0.05), compared with those in the control group. These indexes were reversed after the treatment of combination of Danshen root and Milkvetch root injection, and fetal growth and development in treated group were near to those in the normal control group. 5.The obvious placental morphologic change occurred in rats with FGR-induced by smoking and drinking. Combination of Danshen root and Milkvetch root can release this kind of change.Conclusions 1.The expression of IGF-I and IGF-IR were related to FGR. The effect of combination of Danshen root and Milkvetch root on preventing and treating FGR may be attributed to regulating the expression of IGF-I and IGF-IR directly or indirectly, combination of Danshen root and Milkvetch root can promote fetal growth and development. 2. Smoking and drinking can result in changed structures and characteristics of placental, the pathological changes are the morphological base that lead to the hypofuction of placental. Combination of Danshen root and Milkvetch root can promote the growth of placentals.
