Psoriasis is a dysimmunity, chronic, inflammatory and proliferative skin disease,which is decided by polygene inherited and induced by some environment. No one can illuminate its real cause and pathogenesis till now,but it's also common,relapsing and involve the body areas extensively,so it cause the physical and mental effect significantly in patients. Although the therapic methods are multiformity currently, the therapeutic effects are short and can't prevent them recurrence effectively. The application of some drugs was confined for some reason(such as long-term toxicity,expensive, treatment resistant, and son on).β-carotene is considered to be non-toxic,secure, nutritional food additive by Codex Committee on Food Additive of Food and Agriculture Organization of the United Nation and World Health Organization. To discuss the mechanism ofβ-carotene on treating psoriasis through studying its effect on cell proliferation and cell cycle of Human Umbilical Vein Endothelial Cell(HUVEC) and keratinocyte line aCaT, which thus provides theory support for usingβ-carotene to treat psoriasis.The experiment was divided into three parts: firstly, the primary culture of HUVEC and was identified by means of observing cell shape through inverted microscope and of immunohistochemical staining for factorⅧrelated antigen (FⅧA).Secondly,β-carotene was deliquesced in Tetrahydrofuran(THF), then added into 96 holes plate by certain dilution ratio after 24 hours' culture, MTT was used to detect the optical density(OD) ofβ-carotene on HUVEC and HaCaT after 48 or 72 hours' ??continuing culture. The results show that the effect ofβ-carotene on proliferation of HUVEC has statistical significance(Ρ<0.05) compare with control group when the concentration ofβ-carotene increased to 10-5mol/l, and the effect ofβ-carotene on proliferation of HaCaT has statistical significance(Ρ<0.05) compare with control group when the concentration ofβ-carotene increased to 5×10-5mol/l. Thirdly,choosing one group of being inhibited markly and having statistical significance and analyzing the effect on cell cycle by flow cytometer, the results show thatβ-carotene induced G1-phase arrest of HUVEC and S-phase arrest of HaCaT.Conclusion: The method of HUVEC primary culture is achievable easily and have high success rate, and can meet experiment require.β-carotene can inhibit the proliferation of both HUVEC and HaCaT, and the effect of proliferation become more stronger along with the concentration of drug increasing after arriving at some certain concentration, the mechanism of inhibiting maybe relate to its cell cycle,β-carotene can induce G1-phase arrest of HUVEC, but S-phase arrest of HaCaT. |
