The Study On The Effects Of Papaverine On Flap Survivalafter Local Application In Rats | | Posted on:2008-08-11 | Degree:Master | Type:Thesis | | Country:China | Candidate:H M Hu | Full Text:PDF | | GTID:2144360215488901 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Objective:The aim of this study is to examine the effects of papaverine on flap survival after local application in rats.Method:1.1aboratory animal:60 healthy rats were divided randomly 2 groups and weighed and ordered.2.The creation method of flap:A rectangular random skin flap(3 cm x10 cm)was outlined as on the back of the animals and the cranial side was designed as pedicle.The flap was raised superficial to the deep fascia of muscle and was included the superficial fascia,panniculus carnosus,subcutaneous tissue,and skin.The papaverine or saline was injected under the flap before the skin random flap was cut.In the papaverine injection group before raising the flaps,3.0 mg/kg papaverine was injected to the points defined earlier and was injected under the pedicle for 4 days after operation.the same volume of the saline was injected into control group.4.To draw the materials from the flaps:Full—thickness of the middle flap(n=l0)which size was about lcmxO.5cm was taken respectively at the moment when the flap was created 24 hour later after operation in the control group and the papaverine group.the obtained flap were conserved in a freezer in order to detected the content of malondialdehyde(MDA).Full—thickness of the middle flap (n=10)which size was about 0.3cmxO.3cm was taken respectively in the 4th day after operation was conserved in 4%paraformaldehyde in order to commit paraffin section and dyeing of mmunohisco chemistry.The flap viability in these two groups was observed ten days after the pedicle was cut.5.The observation marker(1)According to the specification of the test kit,the content of MDA were measured.The quantity of the protein in the obtained flap was measured with the test kit of coomassie light blue.(2)In the 10th day after the pedicle was cut, the flap viability was observed.The areas of the total skin flap and the survival skin flap were depicted on a sulfuric acid paper.The papers on behalf of the flap area were sheared and were weighted with the electronic scale.The flap area was reprensented by the paper's weight.Then the flap viability was calculated.(3)The method of image analysis:The skin grafts were sampled to observe numbers of blood capillary and growth of granulation tissues with microscope.6.Analysis of results:Different kinds of results were dealed with by using SAS6.12 software.Results:l Flap survival was 89+4%in the test group and 63+7 % in the control group. There was significantly(p<0.05)difference in the two groups. 2 Microvessel numerber:the microvessel numerber result are shown in the table2 and random skin flap in the test group showed significantly(p<0.05) increased the microvessel numerber when compared with the control group. 3 The level of MDA in the 4th day after operation:Flaps in the test group had a mean level of 0.564+0.245 % VS.1.108+0.237 % in the control group (p<0.05).There were significantly difference between two groups.Conclusion:In conclusion,papaverine is as effective when administered locally into the flap as in the case of systemic administration.By local application,it is possible to avoid unwanted systemic side effects.We conclude that this procedure can be adapted easily to clinical practice, and can be a reliable alternative for increasing flap viability. | | Keywords/Search Tags: | skin flap, papaverine, Cutting pedicle in early stage, survival rate, Microcirculation, ischemia—reperfusion injury, immunohistochemistry | PDF Full Text Request | Related items |
