Objective: Osteoarthritis (OA) is one of the most common diseases in the middle and aged people. It is characterized by articular cartilage degradation, hypertrophic joint change and synovial membrane inflammation. Mediators of inflammation and cytokines play key roles when the disease comes on. There are many ways to treat OA. Most of them are related to antagonistic relation of synthesis and detriment cytokines. This paper is to research the concentrations of NO and PGE2 in the supernatant fluid of rabbit articular chondrocytes induced by IL-1, the mechanisms in the course of OA treatment by IGF-1, and give some evidence from theory and experiment to prevent and cure OA .Methods: Taking the chondrocytes from two months old rabbit articular carriage, the research firstly cultivated the primary chondrocytes and identified them, and then to culture them in plate with or without IGF-1 and IL-1.To absorbe the cell supernant fluid and determine the concentration of NO and PGE2 , the concentration of NO was measured by the ways of nitrate reductase , and PGE2 was measured by enzyme-linked immunosorbent assay(ELISA), and study the correlation between the groups by statistics analysis ways.Results : l.The concentrations of NO and PGE2 in IL-1 10ng/ml group were significantly higher than that from the contral group (P=0.000) .The average value of NO in control was 12.404±8.809μmol/L and its in the first group was 89.971±10.224μmol/L. The average value of PGE2 in control was 1.900±0.227 pg/ml and its in the first group was 12.404±8.809μmol/L (p=0.000) .It was obviously that IL-1 can augment the concentration of NO and PGE2 in the chondrocytes culture supernant fluid in vitro.2 .At the same concentration of IL-l group, The dose-dependent IGF-1 can decrease the concentration of NO and PGE2 in the chondrocytes culture supernant fluid in vitro. The optimization concentration level of IGF-1 was 50 ng/ml. There was statistical significance between the relevant groups.鈽匔onclusion : 1.IGF-1 can decrease the concentration of NO and PGE2 in the chondrocytes culture supernant fluid in vitro.鈽?.It gives some evidence from this experiment that IGF-1 may take part in the cure of OA by decreasing the concentration of NO and PGE2 in OA pathology procedure .
