Histopathology Of Vitreous In Perliferative Vitreoretinopathy And The Expression Of Transforming Growth Factor-β₂ In Vitreous

Objective To detect the histopathological changes in the vitreous of proliferative vitreoretinopathy (PVR) and measure the density of TGF-β₂ in the vitreous of PVR in different grade, and investigate the correlation between the density of TGF-β₂ and the degree of PVR.Methods Observing the histopathological changes in vitreous of PVR and applying Immunohistochemistry method of streptavidin-biotin-peroxidase complex(SABC) to detect the expression of TGF-β₂ in vitreous. Applying enzyme linked immunosorbent assay to measure the density of TGF-β₂ in 30 vitreouses obtained by vitrectomy as well as 5 normal vitreouses, and analyzing the average density of TGF-β₂ in each group of PVR in different grade statisticaly.Results1 Several kinds of cells and collagenous fibers were found in vitreous of PVR. Most cells were epithelial cells and fibroblasts.2 TGF-β₂ was expressed on the epithelial cells and ECM. The density of TGF-β₂ in normal group was 175.91pg/ml. The density of TGF-β₂ in PVR-B group was 257.32pg/ml. The density of TGF-β₂ in PVR-C group was 559.38pg/ml. The density of TGF-β₂ in PVR-D group was 1397.31pg/ml. The difference of the density of TGF-β₂ in each group was significant (P＜0.001). The density of TGF-β₂ and the degree of PVR has linear correlation, r=0.868(P＜0.001).Conclusions The histopathological changes in vitreous of PVR is the production of RPE cells and fibroblasts. There is significant difference between the density of TGF-β₂ in each group. The density of TGF-β₂ in vitreous of PVR is positively related to the grade of PVR.
