A Influence of Da Chai Hu Decoction Reacting to Express of Chemotatic Factor in Aorta of Rabbit with ArtherosclerosisObjective: To explore mechanism of action of Da Chai Hu decoction reacting to Inflammatory reaction in artherosclerosis.Method: Fifty rabbits were randomedly assigned to five groups, including blank, model, xue zhi kang, prophylaxis and treating group, ten cases in each group. Rabbits were fed high-fat forage for ten weeks except blank group to set up AS model. Investigate blood-fat contents in serum by enzymic method when experiment was over; investigate c-reactive protein by immunoturbidimetry when experiment bigins and completes; HE dyeing, to observe information of plaque forming and inflammation infiltrating of artherosclerosis under slight microscope.; to detect the express of mRNA about IL-8 and MCP-1 in aorta by RT-PCR.Rssults:①blood-fat: xue zhi kang group, prophylaxis group and treating group were remarkably lower compared to model group (P<0.01); xue zhi kang group have no influence compared to prophylaxis group(P>0.05); treating group was markedly advanced compared to xue zhi kang group and prophylaxis group (P<0.01).②CRP: there was no significant difference before experiment; CRP level was markedly advanced after experiment besides blank group, xue zhi kang group, prophylaxis group and treating group were markedly lower compared to model group(P<0.01); prophylaxis group was remarkably lower compared to xue zhi kang group(P<0.01); treating group was remarkably lower compared to xue zhi kang group(P<0.05); treating group was remarkably advanced compared to prophylaxis group(P<0.05).③pathematology observing: in xue zhi kang group, prophylaxis group and treating group, the area and thickness atherosclerotic plaque were remarkably littl compared to model group, so did xanthoma cell and inflammatory cell infiltration. there were no necrosis and cicatricle in the atherosclerotic plaque.④Express of mRNA about IL-8: there was almost no express in blank group; xue zhi kang group, prophylaxis group and treating group were remarkably lower compared to model group(P<0.01); prophylaxis group was remarkably lower compared to xue zhi kang group and treating group(P<0.05); there was no significant difference between xue zhi kang group and treating group(P>0.05).⑤Express of mRNA about MCP-1: there was almost no express in blank group; xue zhi kang group, prophylaxis group and treating group were remarkably lower compared to model group(P<0.01); prophylaxis group was remarkably lower compared to xue zhi kang group and treating group(P<0.01); prophylaxis group and xue zhi kang group were remarkably lower compared to treating group(P<0.01). Conclusion:①Da Chai Hu decoction can obviously step down blood-fat and CRP;②Da Chai Hu decoction can obviously delay the emergement of atherosclerotic plaque and the infiltrative of inflammatory cell.③Da Chai Hu decoction can obviously restrain the express of IL-8and MCP-1. |
