【Objective】To study the effects of the expression of bFGF and FGFR after focal cerebral ischemiareperfusion in rats.【Methods】A rat middle cerebral artery occlusion reperfusion (MCAO) model was produced usingthe intraluminal filament method. The rats were divided into three groups: sham operation group (n=4),model control group (n=24) and phycocyanin group (n=24). After MCAO, apoptosis was determinedby situ cell apoptosis detection. The expression of bFGF and FGFR were determined byimmunohistochemical staining.【Results】(1) In the sham operation group and the model control group, there is only a fewbFGF-positive cells were seen in the normal cerebral tissue. In the model control group, theupregulation of bFGF began 6h after ischemia, reached a maximum at 24h, then subsided gradually,but still in high level. In the phycocyanin group, bFGF-positive cells were also mainly in cortex andstriatum, but the number of the cells were significantly more than the number of the model controlgroup. The time-phase pattern of bFGF is similar to the pattern of the model control group. (2) In thesham operation group and the model control group, there is only a few FGFR-positive cells were seenin the normal cerebral tissue. In the model control group, the upregulation of FGFR began 6h afterischemia, reached a maximum at 24h and subsided at 3d, but still in high level. In the phycocyaningroup, FGFR-positive cells were also mainly in cortex and striatum, but the number of the cells weresignificantly more than the number of the model control group. The time-phase pattern of FGFR issimilar to the pattern of the model control group. (3) In the model control group, Apoptosis-positivecells were preferentially located in the penumbral area and progressively increased up to and peaked at24h after reperfusion, and then decreased at 3d, but still in high level. In the phycocyanin group,Apoptosis-positive cells were also mainly in the penumbral area, but the number of the cells weresignificantly low than the number of the model control group. The time-phase pattern ofApoptosis-positive cells is similar to the pattern of the model control group.【Conclusions】The over-expression of bFGF and FGFR might play a key role in ischemic braininjury after MCAO. Phycocyanin can inhibit the over-expression of bFGF and FGFR in the cerebralcortex, and might play an important role in the protection of ischemic neurons...
