Objective To detect the expressions of skp2 and p27kip1 mRNA in endometrioidanenocarcinoma and normal endometrium; to detect their proteins in endometrioidanenocarcinoma, hyperplasia endometriums and normal endometrium.From which to discusstheir roles in the genisis and development of endometrial carcinoma.Methods The expressions of skp2 and p27kip1 mRNA in 37 samples of endometrioidanenocarcinoma and 16 ones of normal endometrial tissue were detected with RT-PCR. The expressionsof their proteins in 37 samples of endometrioid anenocarcinoma and 43 ones of hyperplasiaendometriums as well as 16 samples of normal endometrium were examined withimmunohistochemistry (PV-6000). The relationship between them and clinical pathologycharacteristics was also discussed.Results 1. Skp2 and p27kip1 mRNA: The level of skp2 mRNA expression was 0.79±0.10and 0.16±0.02 in the endometrioid anenocarcinoma tissue and normal endometrium respectively,which showed a stastic significance(t=-31.832, P<0.01).Regard to the clinical pathologycharacteristics, the skp2 mRNA expressed in a higher level in lower grade, lymph nodalmetastases positive and serosal invasion. The expression of skp2 mRNA were irrelevant topatients' age and surgical-pathological stage (P>0.05, respectively). The level of p27kip1 mRNAin endometrioid anenocarcinoma tissue and normal endometrium was no significant differencebetween them (t=1.863, P=0.067).2. Skp2 and p27kip1 protein: The proteins of skp2 were detected in endometrioidanenocarcinoma, hyperplasia endometriums and normal endometriums tissues demonstratedsignificant difference (P<0.001),it is higher in endometrioid anenocarcinoma than the other ones.The expressions of p27kip1 protein in three groups above demonstrated significant difference(P<0.05), it is lower in endometrioid anenocarcinoma than the other ones. Among endometrioidanenocarcinoma group, the expressions of skp2 protein was positively related to pathologicalgrade, myometrial infiltration depth, lymphatic metastasis (P<0.05, respectively); the expressionsof p27kip1 protein was negatively related to pathological grade, myometrial infiltrationdepth,lymphatic metastasis(P<0.05,respectively),but they both were irrelevant to patients' age and surgical-pathological stage(P>0.05,respectively).The expressions of skp2 protein wasnegatively related to that of p27kip1 protein in endometrioid anenocarcinoma(r=-0.408,P=0.012).Conclusions 1. The expressing level of skp2 mRNA and its protein in endometrioidanenocarcinoma was significantly higher; The level of p27kip1 mRNA was not abnormal inendometrioid anenocarcinoma tissue, but the protein of p27kip1 in endometrioidanenocarcinoma was significantly lowerer than that in hyperplasia endometriums and normalendometriums. P27kip1 gene was overexpressed in the protein level but showed no significantchange in the mRNA level in endometrioid anenocarcinoma tissue, which indicated that it wascontrolled in the post-translation level. Skp2 and p27kip1 may play important roles incarcinogenesis and development of endometrioid anenocarcinoma.2 skp2 mRNA and its protein and p27kip1 protein were all strongly associated withpathological grade, myometrial infiltration depth and lymphatic metastasis, which indicated thatthe high expression of skp2 and low expression of p27kip1 may relate with the development ofendometrioid adenocarcinoma.
