Immunohistochemistry was used to assess the expression of COX-2 protein in 50 cases of renal cell carcinoma tissues and 11 cases of normal renal tissues.Results: Positive expression of COX-2 mRNA was detected in 76.00%(38/50) of renal cell carcinoma tissues and in 36.36% (4/11)of nomal tissues by RT-PCR and there was significant difference in epression of COX-2 mRNA between cancer tissues and nomal tissues (P<0.05). Immunohistochemistry showed that COX-2 was expressed in tumor cell and stroma , masculine particular was mainly located in cytoplasm or cellular membrane. It also showed that positive expression ratio of COX-2 protein was 68% (34/50) in renal cell carcinoma tissues and 27.27% (3/11) in nomal tissues and there was significant difference in epression of COX-2 mRNA between cancer tissues and nomal tissues (P<0.05) . However, there was no significant difference between the different pathology (P>0.05) and grade (P>0.05) .There was significant difference in expression of COX-2 protein of different tumor stages (P<0.05)and metastasis(P<0.05). The expression of COX-2 was up-regulated in the higher stage and metastasis of tumor. Conclusions: It is showed that the expression of COX-2 mRNA and protein in carcinoma tissues of the patients with RCC is significantly positive.The expression of COX-2 is correlated with stage, invasion and metastasis. COX-2 maybe plays a certain role in carcinogenesis and progression of renal cell cancinoma.
