| Objectives:It is one of the methods with high therapeutic efficacy for tumor treatments that the vaccine is used as active immunotherapy method to activate an effective anti-tumor immunoreaction in patients with minimal residual tumor, or as inoculation population to prevent tumor. Also, it has been a particularly active area in immunological research field. In this study, preventive and therapeutic effects of MFC tumor cell vaccine modified with curcumol to the tumor-bearing mice were observed, and the mechanisms of these activated therapeutic effects were explored.Methods:Injecting MFC cells into the subcutaneous tissue and tail vein of mice, was performed to establish the mice gastric carcinoma models and experimental lung metastasis models, respectively. After MFC cells were modified respectively with curcumol under 6.25μg/ml,12.5μg/ml and 25μg/ml concentration to structure the novel tumor vaccines. The cytotoxic activity of spleen lymphocytes and the activity of lymphocyte proliferation were tested by MTT colorimetry. IL-2 concentration was measured from serum by enzyme-linked immunosorbent assay. The inhibitive rate of tumor weight and tumor cells lung metastasis was counted. Preventive and therapeutic effects of MFC-tumor vaccines were observed.Results: (1) Stable mice subcutaneous gastric carcinoma models and lung metastasis models were established.(2) Immune prevention of vaccine: All novel tumor vaccines modified with curcumol under 6.25μg/ml,12.5μg/ml and 25μg/ml concentration can restrain the outgrowth of tumor, and increase the cytotoxic activity of spleen lymphocytes and the activity of lymphocyte proliferation, and restrain MFC tumor cells to transfer to lungs, and there are significant differences between the experimental group and the control group with the natural salt (P<0.05). There are no significant differences among three groups of novel tumor vaccines modified with curcumol(P>0.05).(3) Immune therapy of vaccine: All novel tumor vaccines modified with curcumol under 6.25μg/ml,12.5μg/ml and 25μg/ml concentration can increase level of serum IL-2 of the tumor-bearing mice, and restrain the outgrowth of tumor. The necrosis of tumor tissues and infiltration of inflammatory cells were observed pathologically. There are significant differences between the experimental group and the control group with the natural salt (P<0.05). There are no significant differences among three groups of novel tumor vaccines modified with curcumol(P>0.05).Conclusions:(1) The reliable mice gastric carcinoma model and experimental lung metastasis model can be established with subcutaneous injection and tail vein injection.(2) The novel tumor vaccines modified with curcumol under 6.25μg/ml,12.5μg/ml and 25μg/ml concentration have distinctive protective effect against tumor MFC. According to performance-price ratio of drug concentration, curcumol under 6.25μg/ml concentration is best.(3) The novel tumor vaccines modified with curcumol under 6.25μg/ml,12.5μg/ml and 25μg/ml concentration have certain effect on inhibiting mice tumor, and its immune protection against tumor attack is more significant. According to performance -price ratio of drug concentration, curcumol under 6.25μg/ml concentration is best. (4) The novel tumor vaccines modified with curcumol under 6.25μg/ml,12.5μg/ml and 25μg/ml concentration can activate specific cellular immune response and improve the anti-tumor immunity. According to performance-price ratio of drug concentration, curcumol under 6.25μg/ml concentration is best. |
