| Sanguisorba officinalis belonging to Rosaceae Sanguisorba L was aperennial herbaceous plant. It distribute in northwestern, northeastern, northern,southern region and Guangxi in China extensively. The root of sanguisorbar isabundant in tannis, triterpenoid saponin etc which were bioactive components,so it was worthy of developing. The Sanguisorba officinalis root was used as theobject, restraint of hyaluronidase activity was used as a method to appreciate theantiallergic of samples, some experiments were carried out which concludeextracting with aqueous ethanol, separating and purifying with DiaoionHP-20,hyperfiltration membrane, polyamide and Sephadex LH-20.Thedetection experiments were carried out at the end of separation which concludethin layer chromatography, tube testing, UV and IR. The oxygen free radicalsscavenging test were carried out on the extracting samples in the same time, theaim was to evaluate the antiallergic mechanism of Sanguisorba officinalissample. The results were showed that:The best extracting conditions were chosen as: 60% aqueous ethanol, 50℃,sample weight to solvent volume: 1/40, 1hour/time, 4times. Macrospore resin had better absorbance ability to the antiallergic material, so it can separate theantiallergic material well, the highest antiallergy properties of the componentseparated by HP-20 was 76%. The sampleâ… was separated and purified withhyperfiltration membrane, polyamide and Sephadex LH-20, the inhibition ratioof hyaluronidase was 89%(2mg·mL-1),it was the best inhibition component ofSanguisorba officinalis. The sampleâ… turned blue when touchingFeCL3,turned orange when touching anisaldehyde-sulfuric acid. It can depositedgelatin and bromine water. These phenomenons showed that it belong tocondensed tannins. There were polyphone's characters on UV and IR spectrum。At the same time, the radicals scavenging effect of extraction fromSanguisorba officinalis was higher, the Sanguisorba officinalis sample purifiedwith macrospore resin HP-20 eluted with aqueous-ethanol(60:40,v/v) wassimilar to procyanidins, the scavenging ratio of Hydroxyl free radical, hydrogenperoxide and superoxide anion free radical(·O2-) was 98%, 73% and 45%respectively. Contrasting the dates, there was a positive relativity betweenanti-allergic and anti-oxidation activities. It implied that the antiallergicmechanism of Sanguisorba officinalis sample was related to the scavengingratio of oxygen free radicals. |
