Silk fibroin is a natural protein used as textile material for a long time, consists of 18 kinds of amino acid and has excellent physicochemical characters and superior biocompatibility. These make silk fibroin membrane has superiority and foreground in biomedicine material. But pure silk fibroin film is frangibility and can be dissolved in water easily. Chitosan is a natural macromolecule compound and has unique chemical structure. Chitosan membrane has better physical mechanism property and favorable water absorption.In order to improve silk fibroin membrane's capability, we use chitosan,glutaraldehyde,glycerol and PEG to blend with silk fibroin, partly prepare silk/ chitosan/ glutaraldehyde and silk/chitosan/PEG blend membrane. Discussing the effort of condition of preparation to membrane's structure and capability, ascertain best preparation technics and study blend membrane's cell compatibility. It affords testify for silk fibroin material in biomesicine. The main results are as follows:1. Blend ratio, temperature, glutaraldehyde and glycerin of silk fibroin membrane have remarkable effort to blend membrane physical capability. The degree of swelling rate, water solubility, breathe freely rate, modulus, relative break stress and relative break strain increase with the increase of concentrations of chitosan. When the temperature is 40℃, physical capability is good. When the concentration of glutaraldehde is 0.3%, it is a turning. With the increase of glycerin, physical capabilities have current of reduce.2. Blend ratio, the concentration of PEG and interaction effort of silk fibroin membrane have remarkable effort to blend membrane physical capability. The degree of swelling rate, water solubility, breathe freely rate, modulus, relative break stress and relative break strain have an increasing trend with the increase of concentrations of chitosan. A turning point of physical properties is 0.5% concentr- ation of PEG. 3. Transect of pure silk fibroin membrane is dense and uniform and transect of pure chitosan membrane is exhibit roughness. The transect of blend membrane of 55% silk fibroin , 45 % chitosan , 0.3% glutaraldehyde and glycerin have evidence reticular folding. And the transect of blend membrane of 70% silk fibroin , 30 % chitosan , 0.3% glutaraldehyde and glycerin is smooth ,only have process. The surface of pure silk fibroin membrane, blend membrane of 70% silk fibroin, 30 % chitosan, 0.3% glutaraldehyde and glycerin, blend membrane of 55% silk fibroin, 45 % chitosan, 0.5% PEG and blend membrane of 85% silk fibroin, 45 % chitosan, 0.7% PEG is smooth and the surface of blend membrane of 55% silk fibroin, 45 % chitosan, 0.3% glutaraldehyde and glycerin is exhibit roughness.4. We found that silk fibroin and chitosan just is physical blend by infrared spectrum comparison, and they were compatible with each other. Adding glutaraldehyde make absorption peak in 1546 move high wave band and absorption intensity is enhance. Glycerin has no effort on the structure of blend membrane. It indicates that PEG turns the structure of blend membrane intoβ-sheet.5. We confirmed the combine conditions of composite: 1.silk fibroin content is 70% or 55%, temperature is 40℃, both of the concentrations of glutaraldehyde and glycerol are 0.3%.2.silk fibroin content is 70% or 55%, the concentrations of PEG are 0.3%.3.silk fibroin content is 85% and the content of chitosan is 15, the concentrations of PEG are 0.3%.6. Cell cytoxicity of silk blend membrane by MTT method is 0 or 1 grade when it is 2,4,7d. Cell proliferation is between 85-105% and attachment ratio is more than 90% at 24 hour. Silk blend membrane of test obtain application requirement of biological material.
