| Objective Through determining HBV genotypes of 132 patients and analysing thedistribation of genotypes,to explore the role of IL-18 and the relationship between genotypeand serum level of IL-18 of patients of chronic hepatitis B virus Infection.Methods This test is a genic analysis of serum DNA of 132 cases of chronic hepatitis Bvirus infection by PCR with sequence-specific primers (PCR-SSP). Serum levelof IL-18 of132 patients was measured by ELISA,and 22 normal people were as controls .Therelationship between genotype,condition of disease, hepatic function, quantity of HBVDNA,HBeAg and Serum level of IL-18 were analysed.Results Among the 132 patients,the positive rate of type B,C,B/C and non-A-F were31.81%, 62.13%, 2.27% and 3.79% respectively. Genotype C of patients of chronic hepatitisB virus infection. was predominant.The distribution of genotype B of chronic asymptomaticcarrier was higher than chronic hepatitis B and hepatocirrhosis ,however, the distribution ofgenotype C was lower than chronic hepatitis B and hepatocirrhosis ( Chi Squaer Test orFisher Exact Test,P<0.01 ). The distribution of HBeAg positive patients and HBeAgnegative patients was no. significant difference between genotype B and genotype C (ChiSquaer Test, x~2=0.001,P>0.05) . Serum level of IL-18 of normal controls was significant lowthan patients of genotype B and genotype C ( H Test, H=33.112,P<0.01 ) ,however, therewas no significant difference between patients of genotype B and patients with genotype C (P >0.05) . Serum level of IL-18 was no significant difference between high quantity ofHBVDNA and low quantity of HBVDNA(zTest, z=-0.521,P>0.05) . Serum level of IL-18of patients of chronic hepatitis B virus infection was correlated with ALT,AST and TBIL,andr_swere0.237,0.271and 0.259 respectively(Spearman Rank Correlation Test, P<0.05 ) .Serum level of IL-18 of patients of chronic hepatitis B virus infection was significant different(HTest, H=53.874,P<0.01 ) .The order was normal controls 0.05) . Serum level of IL-18 wasno correlation with quantity of HBeAg and HBsAg(Spearman Rank Correlation Test,P>0.05) .Conclusion Among 132 patients ,there were genotype B, genotype C and genotypeB/C. Genotype C was predominant. The distribution of genotype B of chronic asymptomaticcarrier was higher than chronic hepatitis B and hepatocirrhosis ,however, the distribution ofgenotype C was lower than chronic hepatitis B and hepatocirrhosis.There was no differencebetween HBeAg positive patients and HBeAg negative patients. Serum level of IL-18 ofnormal controls was significant lower than patients of chronic hepatitis B. Serum level ofIL-18 of patients of chronic hepatitis B was correlated with inflammation of liver andcondition of disease. Serum level of IL-18 was no correlation with genotype, quantity ofHBVDNA and HBeAg. |
