Objective: To observe the effect of exogenous recombinant human TNF-a on the P,E2,β-HCG,supernatant glucose,generation and GLUT1,IRS-1 of early human trophoblasts cell in vitro; Insulin signal protein expresses in early trophocyte cell.Methods: The effect of recombinant human TNF-a on secretion of E2,P,β-HCG,resorption of glucose and expression of Gult1,IRS-1 protein in human trophoblasts cell was investigated with Isotope,Biochemistry and Western Blot; The effect of recombinant human TNF-a on generation of human trophoblasts cell in vitro was investigated with MTT; Expression of GLUT1,IRS-1,ERK,GLUT4,CK7,Vimentin in human trophoblasts cell was investigated with immunity class; The effect of 20000pg/ml recombinant human tumor necrosis factor alpha and insulin on resorption of glucose was investigated with biochemistry.Results: 1. The Purity of the early trophocyte cell is high2.GLUT1,IRS-1,ERK,GLUT4,CK7 express in human trophoblasts cell; Vimentin doesn't express in human trophoblasts cell; 3.The effection of recombinant human TNF-a on secretion of P,β-HCG, resorption of glucose have any statistics differences, but P go up with concentration of TNF-a; Result of E2: 200pg/ml,2000pg/ml,20000pg/ml group compare with blank group have statistics differences (p<0.05);4.The effect of20000pg/mlrecombinant human TNF-a and 100nm/l insulin on resorption of glucose of human trophoblasts cell have any statistics differences, but resorption glucose of 20000pg/ml recombinant human TNF-a and 100nm/l insulin group is very low; 5.The effect of recombinant human TNF-a on IRS-1 protein in human trophoblasts cell is completely blocked ;GLUT1 protein hasn't statistics difference; 6. TNF-a can decrease quantity of human trophoblasts cell, but among of TNF-group have any statistics differences;Conclusion: 1.GLUT1,IRS-1,ERK,GLUT4,CK7 express in human trophoblasts cell; 2. TNF-a can decrease quantity of human trophoblasts cell; 3. TNF-a and P may bring about IR; 4. 20000pg/ml recombinant human TNF-a and 100nm/l insulin increase resorption glucose into human trophoblasts cello 5.The effect of recombinant human TNF-a on IRS-1 protein in human trophoblasts cell is completely blocked.
