OBJECTIVE:In present study, the aim was to investigation the effect of total favonoids of Padix Hedysari on K562 cells proliferation, cell cycle, apoptosis and relative gene expression was further determined in an attempt to provide a pharmacological basis for future clinical evaluation.METHODS:1. The effect of total favonoids of Padix Hedysari on K562 cell line survival was deter-mined using the MTT reduction assay. The time and dose-dependent manner was also observed.2. Cell cycle and apoptosis was analyzed by flow cytometry.3. The immunocytochemistry method was applied to quantitative analysis the effects of flavonoids of Radix Hedysari on changes P21 and PCNA gene expression.RESULTS:1. Flavonoids of Radix Hedysari (20~100μg/ml) significantly inhibited the proliferation of K562 cells in a time-and concentration dependent manner.2. After the K562 cells cultured for 48 hours, total flavonoids of Radix Hedysari had no effect on apoptosis of K562 cells but showed significantly inhibition(P<0.01), which was showed that total flavonoids of Radix Hedysari could induce K562 cells arrested at G0/G1 and G2/M phases.3. Compared with the control group, P21 expression was increased, and PCNA gene expression was decreased significantly in K562 cells treated with total flavonoids of Radix Hedysari (40μg/ml,80μg/ml) for 48h.CONCLUSION: 1. The inhibitory effect on proliferation of K562 cells was observed in groups treated with flavonoids of Radix Hedysari in a time-and concentration dependent manner.2. Total flavonoids of Radix Hedysari had no effect on apoptosis of K562 cells but showed significantly inhibition(P<0.01), which was showed that total flavonoids of Radix Hedysari could induce K562 cells arrested at G0/G1 and G2/M phases.3. P21 expression was increased, and PCNA gene expression was decreased significantly after K562 cells treated with flavonoids of Radix Hedysari. |
