To establish the basis for studying on the anti-tumor effect of Trichinella spiralis. We found that the growth of A549 tumor cell in Balb/c mice was inhibited by the antigen of Trichinella spiralis. The antigens of Trichinella spiralis and A 549 tumor cell were found to have fos-related antigen by enzyme linked immunosorbent assay (ELISA) preliminary. The molecular weight of fos-related antigen was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and Western blotting. The location of the fos-related antigen between Trichinella spiralis and A549 lung cancer cell were determined by immunohistochemistry.To study the anti-tumor effect of Trichinella spiralis on A549 lung cancer cell, we vaccinate Balb/c mice with weakened and normal Trichinella spiralis then transplant A549 lung cancer cell to Balb/c mice. The growth inhibition effect of Trichinella spiralis on A549 lung cancer cell in Balb/c mice was studied through morphological detection of tumor in mice, analysis of the mice mortality and the ratio of inhibited tumor. The anti-tumor mechanism of Trichinella spiralis was discussed by determination of immunological index. Compared with control group, the tumor volume and weight in normal Trichinella spiralis vaccinated, ultraviolet weakened Trichinella spiralis vaccinated and Co60 weakened Trichinella spiralis vaccinated mice were significantly decreased. The anti-tumor effect of normal Trichinella spiralis is higher than that of weakened Trichinella spiralis. The higher ratios of CD4+ to CD8+ in three vaccinated groups than that of control group indicated that Trichinella spiralis has overt inhibitory effect on the growth of A549 lung cancer cell. The effect of different vaccination time on tumor inhibition was analyzed and we found that the tumor volume and weight in lung cancer mice vaccinated for 11 days were lower that that of lung cancer mice 7 days before vaccination. Compared with the lung cancer mice of 7 days before vaccination, the mice of 11 days after vaccination have better anti-tumor effect.Balb/c mice were vaccinated with Trichinella spiralis antigen and transplanted with A549 lung cancer cell. The morphous of tumor in mice , the mortality of mice and the ratio of inhibited tumor were studied to observe the growth inhibitory effect of Trichinella spiralis antigen on A549 lung cancer cell. We found that the tumor volume and weight of mice vaccinated with Trichinella spiralis ES antigen were smaller than that of mice vaccinated with Trichinella spiralis soluble coarse antigen. The tumor volume and weight of the mice of the above two groups were significantly lower than that of mice vaccinated with immunoadjuvant. The ratio of CD4+ to CD8+ was significantly higher in Trichinella spiralis ES antigen vaccinated mice than that in Trichinella spiralis soluble coarse antigen vaccinated mice. The results indicate that every form of Trichinella spiralis antigen has anti-tumor effect and Trichinella spiralis ES antigen was better than whole body antigen in tumor inhibition. ELISA was used to determined whether A549 lung cancer antigen has cross-reaction with serum immunized with Trichinella spiralis and Trichinella spiralis antigen has cross-reaction with serum immunized with A549 lung cancer cell. The operation of ELISA was established according to basic procedure. We found that Trichinella spiralis larva in muscle and A549 lung cancer cell have fos-related antigen that has cross-reaction. That was beneficial to determine the molecular weight of fos-related antigen.The components of A549 lung cancer cell antigen and Trichinella spiralis antigens were separated by SDS-PAGE. Western-blotting was used to further determine the molecular weight of fos-related antigen. Three pairs of protein zones with similar molecular weight were found to exist in both A549 lung cancer cell antigen and Trichinella spiralis antigen. The molecular weights of three pairs of protein zones were 97.4kDa, 68kDa and 46kDa separately. The foundation to determine the amino acid sequence of fos-related antigen was established.The location of fos-related antigen was determined by immunohisto -chemistry. The trial was carried out according to the operating instruction of the kits. We found that the antibody of fos-related antigen can cohere to the surface of A549 lung cancer cell. That could be the basis to study the anti-tumor mechanism of Trichinella spiralis.
