With the development of science and technology, the nuclear energy and technique are extensively applied in many fields, such as industry,agriculture, medical and life science and aerospace science,etc. The opportunity which people getting in touch with radiation increases day by day, the possibility of suffering radiation harm increases immediately. The radiation is a kind of serious harmful factor to human body health. That body receives a large dose of radiation can cause acute radiation disease in body such as hematopoietic function barrier, the gastro-intestinal tract damage or the brain damage as the basic pathological changes. Receiving a long time radiation of small dose will appear the symptom of the hematopoietic system damages primarily, accompanied by other system disease; the damage of the radiation even has the far effect and the carcinogenic effect to the body. At present,chemical drugs radioprotected mainly in clinical application have higher effects,but also higher adverse reaction and drug resistances. Consequently, natural medicine with its advantages of small poisonous effect and the rich resources is quickly becoming new hotspots as radiating protection medicine, which has already been valued biggest in the modern medicine health fields.Alcohol Extract of Bark of Juglans Mandshurica Maxim is the medicine in this research. The purpose of this study is to investigate the protective effect of AEBJ against damages of hematopoietic system,immune system and anti-oxidant system induced by radiation and the mechanism involved, and first report the experimental evidences for application of AEBJ as an anti-radiation agent. 1,Qualitative analysis on the components of AEBJ: Chemical method and TLC were used to identify AEBJ. The experimental result indicates that the main components are flavonoids,saponins,anthraquinones, phenols and tannis. 2,Toxicity of AEBJ in vivo: The maximum dose for mice was determined by the acute toxicity test, acute-toxicity experiment showed that the largest i.g amount of AEBJ were 15g/kg in mice. AEBJ is a nonpoisonous substance. 3,Antioxidation of AEBJ in vitro: The hemolysis of red blood cells by itself and H2O2 were measured by colorimetric assay. AEBJ is able to inhibit hydroperoxide-induced oxidation and auto-oxidation of RBC.4,Anti-radiation effect of AEBJ in vivo: Mice were exposed to total body radiation with a single dose of 2.0 Gy X-ray. We observed the effects of AEBJ on the damage of hematopoietic functions, immune functions and anti-oxidant system. Examine the damage of the hematopoietic system including the number of WBC, the number of LYMPH, LYMPH%, the number of bone marrow cells, the quantity of Fe, cell cycle of bone marrow; the immune system including viscera index, ability of lymphocyte transformation and the concentration of MDA and H2O2 of the anti-oxidant system. AEBJ can repair the decreasing of WBC, LYMPH,LYMPH% and bone marrow cells; AEBJ can repair the raising of the quantity of Fe; AEBJ can repair the confusion of cell cycle of bone marrow.AEBJ can repair the decreasing of viscera index and ability of lymphocyte transformation. AEBJ can repair the raising of the concentration of MDA and H2O2.Conclusion The main components of AEBJ are flavonoids, saponins, anthraquinones, phenols and tannis. AEBJ is a nonpoisonous substance. AEBJ is able to inhibit hydroperoxide-induced oxidation and auto-oxidation of RBC. AEBJ can decrease the damage in mice induced by ionizing radiation in vivo, and AEBJ has the protective effect on immune functions, hematopoietic functions and anti-oxidant system.
