Objective: To observe the therapeutic efficacy of bone marrow stem cell mobilization with rhG-CSF to rat in acute myocardial infarction, and to certificate the selectivity in stem cells homing ischemic myocardium and short term security through comparing the cordis, hepar, lung and skeletal muscle mobilized and not mobilized.Methed: Left anterior cescending coronary arteries were ligated to produce acute myocardial infarction model in SD rats. The rats were divided in to 3 groups: AMI group (n=20), mobilized group (n=21), sham operated group (n=20). Mobilized group rats were mobilized by rhG-CSF, others were given normal sodium. 7 days later, the WBC and MNC in peripheral blood were counted to testify the effect of the mobilization. And the Hearts, lung, hepar and skeletal muscle were harvested at one week and four weeks after AMI modeling for histopathological examination. Immunohistochemisty, HE stain were used to detect infiltration of CD34+ cells in the organs. The heart function and infracted range were measured at four weeks after AMI modeling.Results: One week later, compared with AMI group, the WBC and MNC in peripheral blood increased obviously in mobilized group(P<0.05); and there were a great number of monocytes infiltrating with CD34 expression by immunohistochemisty in myocardial infracted zone in mobilized rats, while less in AMI group (P<0.05).Four weeks later, there was no CD34 expression in both mobilized group and AMI group. The lung, hepar and skeletal muscle in 3 group had no obvious differences all the time. Compared with AMI group, the rats in mobilized group had better heart function (P<0.05) and less infracted range (P<0.05).Conclusions: The rhG-CSF can mobilize bone marrow stem cells to home to infracted zones and differentiate to cardiomyocyte -like cells and capillary endothelium cells. And the mobilization will not effect the lung, hepar and skeletal muscle short term.
