Chinese Traditional Medicine (TCM) is the specific personalization of Chinese medical science, it has ancient origin and long development. As an important part of TCM, cut crude drugs are widely used in medicamentarius, hospital pharmacy and praeparatum processing. But the parameters of cut crude drugs processing procedure are lack of unified quantization standard, which cause to different places have different preparation methods and directly influences on the safety and efficacy of clinical medication.Rhizoma Alismatis Orientalitis are the dried tuber of Alisma orientalis (Sam.) Juzep. They are the kind of TCM in common use. They are described in Shengnong's Herbal fistly and considered to be high grade. They have been used as a diuretic action and are believed to have effect of expelling dampness. Pharmacopoeia of the People's Republic of China(2005 edition) records two kinds of cut crude drugs, raw film of Alisma orientalis and Stir-baked Alisma orientalis with salt solution, the Stir-baked Alisma orientalis with salt solution which can cite drug downlink, enhance nourishing yin, clearing heat and diuretic. However, Pharmacopeia has not developed processing technology to quantify the specific parameters, so the quality of durgs after processing is difficult to control, the safety of clinical medication will be affected.Against the phenomenon mentioned above, we carry out the following studies:1.Based on studying the concocted literature of Alismatis Orientalitis, the L9 (34) orthogonal design was employed to optimize the processing procedure by determine the content of alisol A 24-acetate, alisol B 23-acetate and volatile components. We obtained the optimum process is: add 2 g salt with four times the quality of water per 100 g net herbs, moisten them for 10 min, roast at 70~80℃C for 10 min, remove them from the container and cool down.2.As Alismatis Orientalitis mainly contain triterpenes which don't have conjugated double bond in their molecular structure, we make use of evaporative light scattering detector (ELSD) which is a universal detector to determine alisol A 24-acetate and alisol B 23-acetate contained in Alismatis Orientalitis simultaneously, and study the content changes before and after the manufacture processing. The results showed that these two ingredients in most of raw film are higher than in medical material samples, and the content of alisol B 23-acetate in Stir-baked Alisma orientalis with salt solution is higher than in medical material and raw film. However, the content of alisol A 24-acetate shows great difference among all samples.3.Because of the volatile characteristics of sesquiterpene compounds in Alismatis Orientalitis, we employed steam distillation to extract the volatile components, established capillary gas chromatography (GC) fingerprint of Alisma orientalis and chose 18 peakes as the common peaks. Excel and SPSS software was used for data processing. The results showed that GC fingerprint can reflect the whole picture of volatile components, different sources of Alismatis Orientalitis samples have some differences, the same source of medical material, raw film and the Stir-baked Alisma orientalis with salt solution present all of the common peaks. After concocted the relative peak area of 15 common peaks have increased in varying degrees. The GC fingerprint established can be used to identify the habits of Alisma orientalis herb, but can not discriminate the habit of cut crude drug.Under the guidance of theory of Traditional Chinese Medicine, the subject apply the traditional and modern methods, optimize the Stir-baked Alisma orientalis with salt solution processing technology, combine the determination of effective constituents and the study of fingerprint to study the changes of chemical component before and after the manufacture processing, it can provide scientific reference for establishing concocted norms, studying concocted mechanism and evaluating the quality.
