| Objective:To investigate the expression and significance of transforming growth factorβ1(TGF-β1) , matrix metalloproteinase 9 (MMP-9) in lens epithelial cells(LECs) of Streptozotocin-diabetic rats.To study the effects of TGF-β1 and MMP9 on rats with diabetic cataract,and the relationship between TGF-β1 and MMP-9.Method:30 SD rats were randomly divided into experimental and control groups.rats of experimental group were given 2﹪STZ intraperitoneal injection(55mg/kg in 0.1mol/L sodium citrate buffer).Lens were examined using a slitlamp once a week.At the end of 2 weeks,4 weeks,8 weeks after STZ injection,Lens were isolated.Immunohistochemical streptavidin-peroxidases(SP) method was used to detect the expression of MMP-9 and TGF-β1 in experimental and control lens.Transmission eletron microscope was used to observed lens epithelial cells of experimental and control groups.Results:1 The lens of control group were always completely transparent throughout the period of experiment,and the lens of experimental group began to exhibited vacuolar stage by week 2 and folccule by week 4,and nearly opacity by week 8.2 The expression of MMP-9,TGF-β1were significantly different between experimental and control LECs(P<0.05).3 During the progression of catatact,the expression of MMP-9,TGF-β1 gradually increased(P<0.05).4 expression of MMP-9 was positively correlated with that of TGF-β1(r=0.937,P<0.05).5 In control group lens,the cellular structure in cortex and the junctions between fiber cells were normal;in experimental group lens,the irregular arrange of lens fibers and the abnormal junctions between fiber cells were observed.Conclusion:The expression of MM-P9,TGF-β1 continually increased ,and the expression of MMP-9 was positively correlated with that of TGF-β1,which indicated that the two protein played a critical role during the development of diabetic cataract . |
