Objective: To determine effects of Cur on proliferation and metastasis in ovarian carcinoma cell lines HO-8910.Methods:â‘ Ovarian carcinoma cell lines HO-8910 was cultured in vitro.â‘¡MTT method was applied to measuring the growing inhibiting rates of ovarian carcinoma cell lines, treated with curcumin in different concentration (0,5,10,20,30,40,50,60 umol/l) for 24-96h.â‘¢The morphologic changes of ovarian carcinoma cell lines in vitro treated with curcumin in different concentration(0,5,10,20,30,40,50,60umol/l) for 24h were observed by light microscopes.â‘£The expression value of MMP-2,MMP-9 and TIMP-2 of different group treated with curcumin in different concentration (0,1,5,10,20umol/l) for 24h were determined by immunocytochemis- try ,MMP-2 and MMP-9 mRNA by RT-PCR.Results:â‘ Light microscopic observation showed that the abnormal morphologic changes of ovarian carcinoma cells were obvious in the therapeutic group, compared with the contral group.â‘¡The value of MTT showed that curcumin could effectively inhibit the in vitro growth of human ovarian carcinoma cell lines HO-8910 in time- and dose-dependent manners in 72h.The growing inhibiting rate reached the summit and began to decrease in 96h. The growing inhibiting rate of ovarian carcinoma cell lines treated with curcumin in concentration of 40umol/l for 24h was 51.75%.â‘¢C ompared with that of contral group,the value of MMP-2,9and MMP-2,9mRNA of the therapeutic group was significantly lower (P<0.05). the value of TIMP-2 of the therapeutic group was significantly higher (P<0.05).Conclusion: curcumin could effectively inhibit the in vitro growth of human ovarian carcinoma cell lines HO-8910 in time- and dose-dependent manners in 72h.Curcumin can inhibit the expression of MMP-2,9 and enhance the expression of TIMP-2.These show that curcumin can reduce the expression of MMP-2,9 and regulate the balance of MMPs and TIMPs.It may have some influence on invasion and metastasis of ovarian carcinoma. |
