| ObjectiveTo investigate FOS protein expression in the Medullary visceral zone of the rats following Cerebral hemorrhage(CH) complicated by multiple organ dysfunction syndrome (MODS); To discuss the possible pathogenesis of Medullary visceral zone in the rats following CH complicated by MODS.Method1.Model of Cerebral hemorrhage was induced in rats by injecting Collagenase VII into the caudase putamen.80 Wister rats were randomlydivided into four groups: normal control group(n=10) , sham-operative group(n=10), Cerebral hemorrhage group(n=30)including 24h, 36h, 48hthree time points, Cerebral hemorrhage+shut off vagus nerve group(n=30)including24h, 36h, 48hthree time points. Incidences of systemic inflammatory responsesyndrome (SIRS) and MODS were diagnosed by their criteria.2.Changes of symptoms and signs including temperature, heart rate and respiration of each group were recorded, the amount of WBC and function of liver and kidney were assayed, the dose of plasma endotoxin were examined. Pathological changes of brain\lung\liver\intestines and kidney were 3.recorded and pictures were taken through optical microscope.The area density of positive staining expressing FOS protein in Medullary visceral zone was analyzed for the relative content of immunohistochemistry.4.Results were expressed as x±s. One-way-anova of SPSS 10.0 soft ware was used to evaluate the difference among groups. Correlation analysis of SPSS 10.0 were used, too.Results1 The rats of hemorrhage group had higher temperature and respiration than nomal group , and control group and sham-operative group (P <0.05) ,and the levels of ALT, AST, BUN, Cr, CK and LDH in those group improved significantly. (P <0.05) There were significant differences between cerebral hemorrhage+shut off vagus nerve group and normal control group, sham-operative group ,shut off vagus nerve group and hemorrhage group (P <0.01 ) , between hemorrhage + vagus nerve stimulate group andhemorrhage+shut off vagus nerve group (P <0.05)2.In hemorrhage group, hemorrhage+shut off vagus nerve group and hemorrhage + vagus nerve stimulate group. there are various inflammatory lesions on the organ tissue at every time point , in hemorrhage group the organ has significantly pathological changes at 24-36 hour point, the changes gradually extinct in 48 hours, and it almost recovers normal at 72 hour point; in hemorrhage+shut off vagus nerve group the inflammatory reaction on lung and small intestine can be seen 4 hours after bleeding, after 12 hours nonspecific inflammatory reaction can be seen on lung\ intestines\liver and kidney, the pathological changes become significant at 24-48 hour point, and there are still significantly inflammatory reaction at 72 hour. hemorrhage + vagus nerve stimulate group ,the organ has significantly pathological changes at 24-36 hour point, the changes graduallyrecovedt in 48 hours.3.The FOS-like neurons were densely expressed in the nucleus of the solitary tract and dorsal nucleus of vagus nerve, and be obviously more than control group , sham-operative group (P<0.01). The expression of FOSprotein in cerebral hemorrhage+shut off vagus nerve group is obviously less than cerebral hemorrhage group. Organs inflammatory of the cerebral hemorrhage+shut off vagus nerve group are more Serious than other group. The expression of FOS protein in hemorrhage + vagus nerve stimulate group is obviously more than cerebral hemorrhage group. and cerebral hemorrhage+shut off vagus nerve group.4.lncidences of systemic inflammatory response syndrome (SIRS) is 100%, the Incidences of MODS of hemorrhage group70%,mortality rate is 40%,the Incidences of MODS of cerebral hemorrhage+shut off vagus nerve group is 75%,the mortality rate is 50%, the Incidences of MODS of hemorrhage + vagus nerve stimulate group is 65%, the mortality rate is50.2%.ConclusionMedullary visceral zone may be one of the direct regulative center after Cerebral hemorrhage by MODS, vagus nerve may be a Bi-directional Channel of the rats following cerebral hemorrhage and multiple organ dysfunction syndrome. |
