The Research Of RhGH On Express Of P53 Gene Of Immune Cells Of Intestinal Wall After Intestinal Ischemia-Reperfusion Injury

OBJECTIVE:To research the influence of recombinant human growth hormone(rhGH) on express of P53 of T lymphocytes and IgA plasmocytes of intestinal wall after intestinal ischemia-reperfusion injury, so as to explore the mechanism of rhGH on immune barrier of intestinal wall.METHOD:60 Wistar rats were divided into two groups randomly. One is experiment group which used rhGH, the other is control group, in addition , each group was divided into three subgroups (2^nd , 4^th , 6^th days, respectively).All the rats were cared in right conditions during the experiment period. Each subgroup has 10 Wistar rats. All rats were made into ischemia-reperfusion model. The rats of experiment group were injected rhGH (1.33u/kg/d) on postmodel everyday. The express of P53 gene of CD₈⁺, CD₄⁺, CD₃⁺ T lymphocytes and IgA plasmocytes of ileal mucosal and laminapropria of the distal end of ileal mucosal tissue were observed by immuohistochemical method,further more, the difference Between experiment group and control group rhGH were compared.RESULT:(1)The express of P53 of CD₄⁺, CD₃⁺ T lymphocytes in control group is higher compared with it in experiment group on the 4^th and 6^th day(P<0.05 P<0.01); The express of P53 of IgA plasmocytes in control group is higher compared with it in experiment group on the 4^th and 6^th day(P<0.05 P<0.01). (2) The express of P53 of CD₈⁺T lymphocytes in control group has no difference compared with it in experiment group (P>0.05) .(3) In control group ,the express of P53 of CD₄⁺T lymphocytes on the 6^th day is higher compared with it on the 4^th and 6^th day(P<0.05); The express of P53 of CD₃⁺T lymphocytes on the 6^th day is higher compared with it on the 4^th and 6^th day(P<0.01 P<0.05),while it is higher on the 4^th day than it on the 2^nd day(P<0.01) ; The express of P53 of IgA plasmocytes on the 6^th day is higher compared with it on the 4^th and 6^th day(P<0.01 P<0.05).(4)In experiment group, the express of P53 of CD₈⁺ T lymphocytes on the 4^th and 6^th day is higher compared with it on the 2^nd day(P<0.05); The express of P53 of CD₄⁺T lymphocytes on the 4^th and 6^th day is lower compared with it on the 2^nd day(P>0.05).CONCLUSION: The express of P53 in gut immune barrier rises during the gut barrier injury, while rhGH can inhabit it and the right use time maybe in 4^th day.Forthermore,rhGH may effect immune barrier of intestinal wall by this means.
