Objective: Many ocular diseases, such as age-related macular degeneration, central serous chorioretinopathy, pathologic myopia disease, diabetic retinopathy, were currently believed related to choroidal circulation disorders. The return of choroidal blood depends on the vortex vein. The disturbance of vortex vein circulation ,may cause many diseases. To explore the relationship between the result of ligation of vortex vein and retinal degenerative diseases , we designed the test ,It may provide valuable theoretical basis for clinical prevention and early treatment of various diseases.Methods: Fifty rabbits were divided into five groups averagely and randomly: control group , group A, group B , group C and group D.Then elevation of IOP in rabbits of group A,B,Cand D wa s achieved by ligating the vortex veins. After treatment ,we observe the changes of eye and measured IOP respectively . The vitreous taken respectively from the rabbits before operation, and 3 days, 1 week, 8 weeks after operation was tested by ELISA for VEGF of rabbit vitreous .And the samples of eyes lweek,8 weeks after operation were taken for HE to watch the pathological ch anges.Result:1.General observation : All time points after eye observation of the model group from both the severity of iris began to congestion, Group D model with the most notable. Group Aand B in 3 hours iris congestion began to alleviate. Model C and D group was in the eyes of both the varying degrees of cornea edema opacity. Group D after 24 h both hyphema, 7-10 days after the basic absorption.2.IOP: IOP in all model groups increased instantly after ligation and model group D was the most prodominant . IOP in model group A increased to(33. 18±3.53)mmHg(1kPa =7. 5 mmHg)after 1 hours and decreased to normal level after 1 day. IOP in model group B increased to (33. 35±2.22)mmHg after 1 hours and decreased to normal level after 1 day. IOP in model group C increased to (45.38±3.99)mmHg after 1 hours and decreased to normal level after 10 days. IOP in model group D increased to (51.90±4.07)mmHg after 1 hours and reached peak, (60.77±4.93) mmHg ,and decreased to (23.94±4.74)mmHg after 6 weeks. Ligation treatment caused a significant IOP increase of the gruoup D in ocular hypertension over group A,B and D(P<0.01).3. VEGF:The level of VEGF in the vitreous of group A and B increased slightly after three days. Since fallen.,normal after 8 weeks .Groups C and D after three days of VEGF were significantly higher, then drop , still higher a week later, Since fallen, normal after 8 weeks.4.Pathology : One week after the operation ,choroidal emerged from varying degrees of congestive, and retinal inner plexiform layer, particle has vacuolar changes, such as nuclear cracking changes, Group D were the most notable. 8 weeks after the retinal layers visible structural disorder, is shrinking kind of change.Surface retinal neovascularization and vascular fiber membranes was found, and internal and external granular layer degeneration, vacuolization, edema, the inner plexiform layer and the ganglion cell layer disappeared in group C and D.Conclusion :1.The vortex vein occlusion can causes retinal degeneration, and provided a theoretical basis to the etiology of the disease related to disturbance choroidal circulation. 2. The vortex vein ligation, to elevate the level of VEGF in the vitreous, may play a role in incidence of ocular neovascular disease.
