Objective: To comprehend the relationship between advanced glycation end products (AGEs) and rat gestation diabetes mellitus (GDM) embryo heart developmental defect, and investigate the mechanisms of the heart developmental defect in infant of diabetic mother (IDM).Methods: To establish GDM group and control one rats model, the SD fem rats were divided into two groups randomly.One was injected streptozotocin(STZ) at belly cavity and the other was injected PBS in the same position.Observe blood glucose and urine glucose every day after 72h when tats were given drugs,All rats were uterine-incision delivery to draw the materials from embryo heart tissues at gestation 13th , 16th, 19th day randomly,heart tissue for pathological change by H.E stain.; The expression of RAGE in heart tissues of all groups were analyzed with immunohistochemical techniques;Heart tissue ultrastructure change at every group by election microscope;Collect artery blood from the mother rats and use the spectrofluorometric method to observe the level of AGEs of the serum. Results: The mother rats of DM group appear the phenomenon of "three more one few".After 3rd day when given drugs,compare to the control group both blood glucose (14.65±1.12vs.3.06±0.39) and urine glucose were higher (p<0.01), weight were less (260.7±14.0vs.273.7±16.7, p<0.01);All abortion rate at DM group were hihger than that of control group (p<0.05),There are obvious difference (p<0.01) at the number of liquefactive embryos, dead ones, huge ones and hypoevolutism ones between two groups. There are more at embryo heart development defect numbers at diabetes group than that of the control group,there exists obvious statistics difference (F=196,P< 0.01),major including caul-conus development malformation ventricular septal defect (VSD),ventricular septal cleft(VSC) and myocardial hypertrophy and so on.The number of AGEs accept fluorescence enhanced follow the duration of pregnancy at mother rat DM group,and were larger than that of control group (p<0.01) and there was obvious differentiate at statistics (p<0.01).
