| Objective To study the expression and position of neuronal nitric oxide synthase(nNOS) and pathological change of muscle fiber in foot muscle of congenital clubfoot (CCF), analyze whether there exists a regular correlation between nNOS and cross section area(CSA) of skeletal muscle fiber, In order to explore the mechanism and significance of nNOS in neuro-muscular etiological theory of CCF and indicate the clinical therapy.Methods We study twenty patients of stiff CCF aged from 6 months to 3 years (mean 18months) requiring surgical treatment. there were twelve boys and eight girls. Muscle specimens of anterior tibial , peroneus brevis, post tibial and gastrocnemius from patients were obtained in the course of corrective orthopaedic surgery of CCF. Foot muscle samples of normal control group from 5 injured children aged 1 to 3 years without neuro-muscular disease in operation. All the specimens were examined using light microscopy and hematoxylin and eosin and anti-nNOS immunohistochemistry staining. We observed the distribution of nNOS positive staining region and pathological change of muscle fiber.Quantitative measurement of the anti-nNOS staining and the CSA of skeletal muscle fiber were performed for statistic analysis and study on association between the expression of nNOS and CSA of skeletal muscle fiber.
