| PURPOSE: The successful enamel formation has something to do with the enamelin gene. Once the gene mutated, the enamel formation will be interfered. Studies have shown that an ENAM mutation causes the local hypoplastic form of ADAI, a phenotype that accounts for 27% of the autosomally inherited cases in Northern Sweden. This mutation in the enamelin gene results in a truncated peptide of 52 amino acids as compared with 1142 amino acids of the normal protein. The purpose of this study was to clone human mutated encoding mutated protein, which provide a basis for expressing the recombinant human mutated enamelin in Escherichia coli. in the future study.METHODS: In this study, total RNA was extracted from the dent germ of a legally aborted embryo, and the synthesis of cDNA was obtained from the total RNA by using RT-PCR technique. Then the desired DNA products were... |
