Influence Of Tanshinoneâ…¡A On Proliferation And Migration Of Rabbit Vascular Smooth Muscle Cells In Vitro

Objectives: 1. To study the influence of Tanshinone â…¡A on proliferation and migration of rabbit vascular smooth muscle cells in vitro; 2. To primarily probe into the mechanism of how Tanshinone â…¡A works. Methods: Rabbit aortic artery was chosen for primary culture of vascular smooth muscle cells (VSMCs). The tissue pieces were cultivated in DMEM with 10% fetal calf serum in vitro. Three weeks later passage was performed. In this experiment the cells of 3-5 passage were used and divided into control group and Tanshinone â…¡A groups according to different concentrations (detailed grouping is in the part of Material and Method). After 24h, 48h, 72h respectively according to the protocal, MTT assay was used to study the effect of Tanshinone â…¡A on the VSMCs proliferation; sterile scraper and contrast phase microscope were used to measure the distance of VSMCs migration in control group and Tanshinone â…¡A groups; Analysis of cell cycle and DNA content were performed by flow cytometry.
