| Background and objective:As the key procedure of tissue engineering ,the gain of seeding cells is very important .The myoblast can be purified from skeletal muscle or induced from MSCs.The present study is to compare the two ways so that conclusion is drawn which way is better for supplying seeding cell of skeletal muscle in engineering tissues. Methods:Trypsinization was employed to culture and purify the myoblast of neomate rats that born in a day,then went down and identified to the future generation.The third generation myoblasts were mixed into cell suspension, cell density of 1×10~5/ml was inoculated in 24 orifices and 96 orifices. Five porocytes were got randomly from 24 orifices in the 1,3,5,7,9d respectively, ten porocytes were got randomly from 96 orifices in the l,3,5,7,9d respectively.Cell viability to 5 porocytes was determined by the analysator of cell viable.While cell viability of the third generation myoblasts to 10 porocytes were determined by MTT, absorbance was measured by 570nm to determine OD of 1,3,5,7,9d,finally drawed cell growth curve of myoblasts by using mean value of cell viability and OD so as to comprehend the state of cell growth.
