Bone diseases such as osteoporosis, osteoarthritisand bone cancer are closely associated with the aging process. Carriers of bone target, such as bisphosphnates, polymalonates, micromolecule heterocycles and peptides were discovered in succession since the first introducing of the concept of bone target by Pierce in 1986.With the fast development of drug design and synthesis of bone target drugs, the experimental pharmacology in this field hardly had great breakthrough except the study of nosogenesis. Around the experimental pharmacology exploratory study of bone target drugs, research works were carried out in aspects as follows:1,After analyzing the limitations of the mainly methods of bone target detection and discussing the numerous factors and complicated relationship in bone target process, the Author introduced two conceptions that bone powder was utilized to take the place of hydroxyapatite and a synthetical detection method was presumpted with bone powder and a simulation solution of bone tissue fluid.2,Two estron derivatives and two isoflavone derivatives were synthesized and their structures were confirmed by UV, ~1HNMR. These four componds were utilized as test materials with another eight tetracycline derivatives (analogues) and four 5-FU-bisphosphonate conjugates in this paper.3,Simulation solution of bone tissue fluid and bone powder were prepared to compare the differents between bone powder and hydroxyapatite in adsorption test in vitro, and the differents between simulation solution of bone tissue fluid and pure solvents.
