Objective: To prepare monoclonal antibody against human lung squamous cancer cells and identify its characterization, so as to help clinical treatment,early diagnosis. It also helps isolate the antigen gene which corresponding to the McAb.Methods: As an antigen human lung squamous cancer cell YTMLC-90 and YTMLC-90 coated by health human lymphocyte was used for immunizing Balb/c mice. McAbs against YTMLC-90 cells was prepared by hybridoma technique---- conventionally fusion mouse myeloma cell SP2/0 cells with spleen cells from Balb/c mice immunized with human lung squamous cancer cell YTMLC-90. Indirect ELISA and limited dilution was used to select hybridomas. The selected hybridoma cells were cultured in vitro and vivo to obtain the cell supernatants and ascites.
