| Objective: Effect of plant Coleus Forskohlii active monomer ISOF and CF-E (analogs of FSK) on ALI in vitro was studied. Moreover, the mechanism about them was investigated primarily. Methods: Effect of ISOF and CF-E on aggregation of PMN activation induced by fMLP performed using a 4-channel platelet aggregometer ; cytometry was adopted to analyze the effect of them on PMN adhension to LPS-activated ECV-304; the expression of PMN adhension molecules was determined with flow cytometry; radioimmunoassay was applied to detect them on the quantity of TNF-a PMN secreted, intracellular cAMP level of PMN was detected by the same way. Results: It was found that ISOF(25,50,100μmol/L) and CF-E(1.25,2.5,5mg/ml) can inhibit PMN aggregation, the maximum agglutination are 43.63 and 55.94 percent; they inhibited PMN adhension to ECV-304 except CF-E(1.25 mg/ml), the maximum inhibition rate are 62.98 and 9.58 percent; ISOF(25,50,100μmol/L) and CF-E(1.25,2.5,5mg/ml) can all inhibit the expression of PMN adhension molecules, the fluorescence intensity of CD lib/CD 18 in the control group are 252.15 ± 12.77, 48.60 ± 0.86, the expression of CD14 is 40.77 ±... |
