Objective: To explore the expression of Survivin characteristics and its effect on breast cancer cell MDA-MB-231 proliferation and their relationship between Survivin,JAK-STAT pathway and HSP90.Methods: MDA-MB-231 cell line was used as the research model, and AG490 as JAK inhibitor and GA as HSP90 inhibitor. Use Western blot to detect the changes of expression of P-STAT3,Survivin and HSP90 protein in the cell. Use RT-PCR to detect the changes of the mRNA of Survivin and HSP90 .Use MTT assay to detect the changes of cell proliferation. Use flow cytometry to detect apoptosis of the cell.Results: JAK inhibitor AG490 reduced the expression of P-STAT3 and Survivin protein and increased the expression of HSP90 protein in human breast cancer cell MDA-MB-231. P-STAT3/β-actin is 0.738±0.018, Survivin/β-actin is 1.096±0.026, HSP90/β-actin is 0.637±0.013 without AG490 and P-STAT3/β-actin is 0.211±0.011, Survivin/β-actin is 0.278±0.012, HSP90/β-actin is 0.815±0.021 after incubation with AG490 for 40μmol/L 24h. At the same time cell proliferation decreased and apoptosis increased. After incubation with AG490 40μmol/L for 24h, the"sub-G1"peak appeared and apoptosis rate is 5.62%; After incubation with AG490 40μmol/L for 48h, apoptosis rate is 28.81%.
