PART ONE EFFECT OF FORSKOLIN ON AQUAPORIN-8 EXPRESSION AND DISTRIBUTION IN WISH CELLSObjective: To study the effect of Forskolin on aquaporin-8 expression and distribution in WISH cells.Methods: WISH cells were divided into control group and experiment group at random. The cells of control group were cultured in normal medium. The cells of experiment group were treated with 5,10, 20, 40, 100, 200μmol/L Forskolin for 24h, respectively. Then treated with 100μmol/L Forskolin for 4h, 8h, 16h, 24h, 48h, respectively. The RT-PCR,Western blot technique were used to quantity AQP8 mRNA and protein expression levels. Immunofluorescence microscopy was used to study the localization of AQP8 in WISH cells.Results: AQP8 protein levels increase in a does-dependent manner in response to treatment with 10~100μmol/L Forskolin.
