Antiviral Activity Of AGM And Its Mechanism In Vitro

PART 1 STUDY OF ANTIVIRAL ACTIVITY OF AGM ON HBV IN HEPG2.2.15 CELLObjective To investigate the antiviral activities to hepatitis B virus(HBV) and cytotoxicity of AGM in HepG2.2.15 cell.Methods 1. After treated with AGM of different concentration (2.5uM ,1.25uM,0.625uM,0.3125uM), 2.2.15 cells were detected the content of HBsAg and HBeAg on supernatant by Abbot kit ,then the inhibitory rate on HBsAg and HBeAg have been calculated. 2. HBV DNA was measured by FQ-PCR. 3. the cytotoxicity on 2.2.15 cells was examined by MTT.Results 1. The inhibitory rate of AGM on HBsAg , HBeAg and HBV DNA was higher than the control group without AGM treatment. The inhibitor rate on HBeAg was much higher than that of HBsAg. 2. There was no difference of the viability of 2.2.15 cells between the two groups with and without AGM treatment by MTT results .Conclusion AGM can inhibit the secretion of HBsAg and HBeAg from HepG2.2.15 cell , and synthesis of HBV DNA.
